Maize dwarf mosaic disease caused by Maize Dwarf Mosaic Virus (MDMV) is one of the most important diseases of maize in the Corn-producing areas of china.Most effective and economical way to prevent MDMV is to cultivate resistant varieties. In this study, we screened out highly resistant and susceptible corns material huangyesi-3, 8112 by identification of resistance to MDMV. The resistant gene was located by BSA(bulked segregant analysis) method using SSR molecular marker, the main results were showed as follows:1)The highly resistant inbred lines huangyesi-3 and the highly susceptible inbred lines 8112 were selected as parents for genetic research from 30 materials. F1 and F2 populations were constructed with the two inbred lines. The analysis of disease-resistance traits of F2 indicated that the resistant phenotype was dominant.2) Among 135 SSR primers, 51 amplified polymorphic primers were detected between two parents, 3 stable polymorphic primers Bnlg1601,Umc1644,Umc1528 were located on chromosome 3 between the resistant and susceptible pools.3)Linkage analysis of 3 primers in F2 populations indicated the Bnlg1601 and Umc1644 linked the resistant gene with the genetic distance 20cM and 14cM, respectively.4) Additional 13 primers in the preliminary location region were selected and screened polymorphically, Genetic Linkage analysis with all polymorphic primers indicate Bnlg1035,Umc2266,Umc1644 were linked with the resistant gene, with the genetic distance 8cM,3cM和6.6cM respectively, and Bnlg1035 and umc2266 were in flank of the resistant gene respectively.In summary, SSR molecular markers linked tightly to the resistant gene were screened and located accurately on the chromosome 3, which provide a theoretical basis for screening the resistant germplasm quickly and cultivating new varieties, and laid the foundations for Molecular marker-assisted breeding and the cloning of resistance gene to Maize dwarf mosaic virus.
