Preparation Of "Qiumie" Granul And Its Effect On Prevention & Treatment Of Coccidiosis

Coccidiosis in chickens belongs to parasitic diseases in the traditional Chinese veterinary area,and to prevent and treat coccidiosis, we use traditional Chinese medicine, to controls coccidiosis.Ji Qiuchong San rooted from the 2005 edition of "The People's Republic of China Veterinary Pharmacopoeia", The Principal constituents of this Proved recipe includes Herba Artemisiae Annuae, Agrimonia Pilosa Ledeb, Pulsatilla chinensis (Bunge) Regel, Polygonum multiflorum Thunb, Cinnamomum cassia Presl, and it has the function of anti-coccidian and haemostasis.To enhance the effects of prevention and treatment of chicken coccidiosis and easily to operate,therefor,e this Proved recipe is changed from the powder into the granule.At last, the granule is named as"Qiumie" granules.This article explored the production processes of "Qiumie" granule.Through the orthogonal test,we studied the best extraction process of the active ingredients from Herba Artemisiae Annuae and theβ-CD inclusion technology of the active ingredients,meanwhile, we selected the optimum extraction process of essential oil from Cinnamomum cassia Presl and theβ-CD inclusion technology of essential oil by orthogonal test; In water extraction craft research of admixture of Agrimonia Pilosa Ledeb, Pulsatilla chinensis (Bunge) Regel, Polygonum multiflorum Thunb, we selected optimum extract conditions with orthogonal design. Subsequently,we studied the ratio of Herbal Extracts and adjuvant in "Qiumie" granules,the concentration of adhesives.By inspecting the color, Sieving rate, yield of the granules,we selected the best molding process,and we investigated the moisture, bulk density, moisture absorption as well as the critical relative humidity of " Qiumie"granules, for large particles mass production and packaging system of the granules.We studied the quality control of the "Qiumie" granules and formulated the draft of the quality standard.We use TCL to identify Herba Artemisiae Annuae, Pulsatilla chinensis (Bunge) Regel, Cinnamomum cassia Presl, with good reproducibility. We determine artemisinin in the "Qiumie" granules by HPLC.The chromatographic conditions:Column:symmetryshield RP 18 (3.9×150mm,5μm), mobile phase: acetonitrile-water (75:25), flow rate:1mL/min The detection wavelength:205nm, column temperature:25℃, sample volume:20μL; The standard curve regression equation:y=204049x+115026 (r=0.9996), the content of artemisinin between 0.2575 and 8.24μg was good. In the experiments of prevention and treatment of chicken coccidiosis,we made chicken coccidiosis with Eimeria coccidia. Comparing with Ji Qiuchongsan and Western medicine,we uesed different doses of the "Qiumie" granules to prevent and treatchicken coccidiosis, so we confirmed the best dose of clinical treatment and prevention of "Qiumie"granules.It is good therapeutic effect to treat chicken coccidia with 0.16% of the "Qiumie" granules through drinking water,and its anticoccidial index (ACI 163.21) is as good as the anticoccidial index (ACI161.34) of Ji Qiuchongsan,so we recommend it is best clinical does of 0.16% for treatment of chicken coccidiosis though water delivery;the anticoccidial index of 0.16% Western medicine "Quanqiutong" (ACI176.17) is similar to the anticoccidial index of 0.24% "Qiumie" granules in high dose group (ACI 178.58) close. In experiment of chicken coccidiosis prevention of Ji Qiuchongsan, it's medium anticoccidial effect,because its ACI is 145.36;It is very good to prevent chicken coccidiosis with 0.12% Qiumie" granule (ACI 175.06),and its is higher than the anticoccidial of 0.03% "Quanqiutong" "(ACI166.07),Therefore,we recommend it is best clinical does of 0.12% Qiumie" granule to prevenchicken coccidiosis though water delivery.
