Litsea cubeba has economic value and Medicinal Value, and it has good use prospects, but the low seed germination influences its application and development。How to improve its reproduction speed is the point that the paper research on。The paper adevelop on experiment of accelerating germination by a series of physical and chemical methods, and it obtained some certain effect。For accelerating reproductive efficiency, this paper also research on tissue culture technology of Litsea cubeba in order to improve its breeding rate。The preliminary research on Litsea cubeba seeds is as follows:Fosterage in warm stratification cannot promote sprouting.During dormancy, the Litsea cubeba seed would go through a low temperature period to complete the internal material transformation to upraise the germination percentage; the germination percentages of the low temperature lamination 0d,30d,60d, and 90d are 1.8%,3.55%,5.45% and 7.63% respectively. Gibberellin poses little effect on promoting germination. The germination percentages of seeds disposed with gibberellin in 0d,30d,60d,90d are 7.57%,7.60% 7.60% and 7.59% respectively. Immersion in mild warm water could promote germination rate effectively. The best condition is immersing seeds 24h in 30℃water, with germination rate 9.83%.Chemical reagents in different concentrations such as niter advance Litsea cubeba seeds'germination rate notably. Niter, borate, IAA, and NAA could advance germination rate, germination condition and percentage, with ranking niter500 mg/L> borate500 mg/L> IAA50 mg/L>NAA100 mg/L.The endosperm is the vital essential part of the seed, in proportion of 82.53%, following by the coat, taking a part of 26.98%. The rest part is embryo,049%. The coat is firm and compact, colored brown。The embryo is tiny; The mature Litsea cubeba seeds have low water content,7.3%, absorption test show that water absorption on speed is slow。The test on internal inhibitor of Litsea cubeba proves The Litsea cubeba has some certain internal inhibito。For the above reasons affect the germination rate of Litsea cubeba seeds and germination speed of germination speed.The preliminary test on Stems with bud of Litsea cubeba seeds is as follows:(1)Different disinfection treatments" effect on Litsea cubeba combination of 0.1% L mercury~+ 15%84 disinfectant poses an ideal effect. The best timing is 10mins~+ 3mins with an average infection rate 17.55%, the lowest one in all. (2)The selection of explantsThe callus rate induction test on explants from top, middle and bottom without buds certifies that the top one pose the highest callus rate,84.44%, following by middle ones.82.22%. and bottom ones.71.11%;stems without bud pose a high contamination rate and lowest callus rate. The top stems with bud is the suitable explants after primary screening(3)The induction test on different hormone concentrations'effect on stem with buds of Litsea cubeba:The best hormone combination of callus induction is MS+6-BA0.7 mg/L+2.4-D0.20 mg/L IBA0.10mg/L.(4)The differentiating tests on Litsea cubeba's callus indicate:6-BA in 0.7mg/L poses a higher callus differentiation rate than other concentrations. What's more.6-BA in 0.7mg/+IBA0.10mg/L poses a differentiation rate at 95.8%. At a comprehensive analysis, the best hormones combinations of stem with bud's callus differentiation are MS+6-BA0.70mg·L-1+IBA0.10mg·L-1.(5) The test on Litsea cubeba's subculture indicatesThe suitable culture medium for Litsea cubeba subculture is 1/2MS+IBA0.05mg/L(6) The radication test on Litsea cubeba indicates:The best rooting culture combination is 1/2MS+30g/L sucrose+0.02mg/LIBA。(7) Acclimatization and transplantsLitsea cubeba's tissue culture seedling needs a transactional stage:First,3days inner natural lighting to adapt to the environment;then, explanting it from the culture room to adapt to the outer environment for 10days.
