Cytoplasmic male sterility(CMS) is useful for theoretical research and agricultural application,however,the mechanism of sterility needs further investigation.Ferritin is an important anti-oxidative protein related to sterility and CMS is accompanied with high oxidative stress.In this paper,we used CMS and MF of mustard(Brassica juncea L.) as materials,and cloned one ferritin gene,and studied characters of ferritin,and from the angle of oxidative stress and anti-oxidative genes expression,we compared the seedings and buds of CMS and MF,which assisted us on understanding the characters of nutritional stage and reproductive stage of CMS and MF.1 We had cloned the total 1085 bp cDNA of ferritin(Bjferritin) in mustard, coding 254 amino acids.We found BjFerritin showed 92%homology with AtFerritinl,and presumed that Bjferritin was Bjferritinl.Using RT-PCR or Real-Time PCR,we found that Bjferritinl had the common property of ferritil(responding to free iron) and the character of ferritinl(responding to H2O2), and didn't have the character of ferritin2(responding to ABA).The results proved that the cloned gene was Bjferritinl,with the characters of ferritinl.2 Using GFP technique,we found BjFerritinl was localized to choroplast, owing to the aggregation of green fluorescence in this organelle.3 We had discovered that both H2O2 content and·OH content in CMS seedings were lower than in MF seedings,which indicated CMS seedings didn't suffer from oxidative stress.By the means of Real-Time PCR,we had found that Bjferritinl,BjCu/ZnSOD(Cu/Zn super oxidative dismutase),BjMDAR1 (monodehydroaseorbate reduetasel),BjAPX8a(ascorbate peroxidase 8a),GR1 (glutathione reduetasel) and BjGR2(glutathione reductase2) were all up-regulated, which indicated anti-oxidative system was activated to prevent CMS seedings from oxidative stress.Although H2O2 content in CMS buds was likewise lower than in MF buds, Bjferritinl,BjAOX1a(alternative Oxidasela),BjCu/ZnSOD,BjMDAR1,BjGR1, BjGR2,BjCAT(Catelase) and BjThioredoxin were all down-regulated.
