PRL which secreted from anterior pituitary gland has extremely broad spectrum of biological role. PRL mainly related to reproduction in avian species. Increased concentration of Prolactin plays a role in the cessation of egg laying and broodiness during the active period of lay. Without exception from the prolactin acceptor adjusts these effects . Large amount of research showed that Prolactin (PRL) physiological action is that the prolactin acceptor (PRLR) is upright by being located in the upper target cytomembrane prolactin lead . In avian, PRLR gene has a very important effect in the development of broodiness behaviour. In this experiment detection of the Wanxi White Geese PRLR gene expression in the pituitary, hypothalamus, ovarian tissue in the a week before laying, 10 days after laying , broody period and maternity leave by real-time PCR . Explored three tissues in different reproductive period of PRLR gene expression.The results showed that PRLR mRNA was expressed in hypothalamus, pituitary and ovarian. The level of PRLR mRNA was expressed from high to low in the three tissues by orders,that is ovarian>hypothalamus>pituitary. the expression of PRLR mRNA between each tissues was significantly different.The expression of ovarian was higher than hypothalamus (P<0.05); the expression of ovarian was higher than pituitary(P<0.01). Analysis of the different periods showed that the expression of PRLR mRNA from high to low were: broody period> a week before laying> 10 days after laying > maternity leave. On the broody period,the PRLR gene expression was 4 times, 40 times, 45 times of that in a week before laying, 10 days after laying and maternity,respectively(P<0.01). The same tissue at different stages of PRLR mRNA expression analysis showed that the pituitary expression of the four periods from high to low were: broody period > maternity leave> 10 days after laying> a week before laying, On the broody period of PRLR gene expression was significantly higher than the other three periods(P<0.01), the other three periods were no significant differences(P>0.05); the hypothalamus expression of the four periods from high to low were: broody period>10 days after laying>a week before laying> maternity leave, on the broody period of PRLR gene expression was significantly higher than the other three periods(P<0.01), the other three periods were no significant differences(P>0.05); the ovarian expression of the four periods from high to low were: broody period>a week before laying>maternity leave>10 days after laying, on the broody period of PRLR gene expression was significantly higher than the other three periods(P<0.01), a week before laying PRLR gene expression was significantly higher than maternity leave, 10 days after laying(P<0.01).In the broody period, PRLR mRNA expression of the three tissues volume were very high, there was no significant difference(P>0.05). The pituitary PRLR mRNA expression in the broody period from high to low were:broody30-31days>broody1-3days>broody14-16days, the broody 30-31days PRLR mRNA expression were 3 times higher than broody14-16days, a significant difference (P<0.05);The hypothalamus and ovarian PRLR mRNA expression in the broody period from high to low were:broody1-3days >broody14-16days> broody 30-31days, no significant differences (P>0.05).
