In this study,SSR markers were developed based on public available genome survey sequences(GSSs) of Brassica oleracea.An SSR linkage map of Brassica napus based on a DH population with 88 lines derived from the No.2127-17 and ZY821 was constructed.The main results are as follows:1.51,088 Genome survey sequences of Brassica oleracea were screened and 60,116 SSRs including dinucleotide to pentanucleotide repeat types,146 kinds of repeat motifs, and repeat length≥11bp were found.Dinucleotide and trinucleotide repeats were dominant,with similar frequency 37.2%and 34.2%in all SSRs,respectively.(AG)n,(AT)n and(AAG)n are the most frequent motifs in Brassica oleracea genome.2.1,398 primer pairs were designed and named BoGMS.Screening the 1398 SSR markers with six Brassica napus lines(S1,S2,M201,M202,No.2127-17 and ZY821), 934 of them successfully amplified PCR products,and 648 performed polymorphism in at least two of the six lines.The trinucleotide repeat type showed the highest successful amplified rates and polymorphism.In view of the frequency in genome and the high polymorphism,(AG)n,(AT)n and(AAG)n motifs were the predominant source of SSR markers developed from genome survey sequences of Brassica oleracea.1398 SSR sequences were compared with the genome of Arabidopsis thaliana.250 sequences had homologous with Arabidopsis,eliminating the results which score<100,E>10-5.3.A genetic linkage map of Brassica napus was constructed using 240 newly developed SSR markers from Brassica oleracea and 198 anchor SSRs based on a DH population.This map contained 496 loci,and covered 19 linkage groups,with a total length 1948.2cM and an average marker interval of 3.9cM.And 33.1%(P<0.01) distorted marker loci was detected in this map.Some of them clustered in linkage groups, and distorted to the same parent.In addition,the most BoGMS marker loci(68.6%) were located on the C genome.
