In this study, total RNA was isolated from tissues of Tibetan and Sichuan Mountainous black-bone chickens(SMBC), Fatty Acid Transport Protein 4(FATP4) gene was proliferated used RT-PCR and the products sequences and amino structure were primary analyzed by bioinformatics. The developmental changes of FATP4 gene expression in SMBC usedβ-actin as internal standard control by the real-time quantitative RT-PCR method , studied the developmental expression of FATP4 gene SMBC We compared the FATP4 gene expression difference in Tibetan, SMBC and Da Heng chicken (DH) . The results are as follows:The coding sequence (CDS) of SMDC chicken FATP4 gene was 1107bp. By analysis of CDS: The homology rates of the nucleotide sequence of SMBC with the original chicken, mice, people, brown rat, is 67.1%, 99.6%, 67.2%, 67.4%, 66.6%和67.3%. The homology rates of amino sequence was 100%, 72.8%, 76.4%, 75.8%和72.8%. The homology rates of the nucleotide sequence and the amino sequence was higher by other species. we have successfully obtained the sequence of SMDC chicken FATP4 gene.Bioinformatics primary analysis suggested that chicken FATP4 has of one possible transmembrane domain and locates in 17-33 spaces of transmembrane protein. The relative molecular weight of chicken FATP4 is 41.47 KDa, and isoelectric point is 7.46.The signal peptide characteristic has found at prediction glycosylation site. It most likely does not have a signal peptide structure. The signal anchor sequence of the existence of a probability is 0.00% at molecular amino acid sequence of chicken FATP4. Exist the possibility of signal peptide sequences is 0.00%.Protein polypeptide chain has a major hydrophobic domain that located in amino acid sequences 9-37, 51-66, 98-115, 128-145 and 250-278. The secondary structure of proteins containα-helices (α-helix, h) 34.78%, an extension of the main chain (extended strand) 16.03%, random coil (random coil) 49.18%.SMBC FATP4 mRNA expression studies have shown differences: 8w, 10w and 12w, respectively, in the PZ, liver and brain tissue with high expression. FATP4: expression in the breast muscle, leg muscle, FZ and the highest values were in the 8w, 8w, 4w and 12w. FATP4: expression in the liver, heart and brain, respectively, the highest value of 8w, 6w, and 8w. Liver is the advantage of expression FATP4 gene. This study found that gene expression changes with time are different. LM, BM, liver and brain expression of the highest is in the 8w; PZ and FZ in the expression levels of the 4w and 12wis the highest respectively. FATP4 gene in the LM, BM, liver and brain expression of the advantages of time is 8 weeks of age;In PZ, FZ and heart of the advantages of expression time points are 4w, 12w and 6w. FZ deposition generally increase with the growth. However, this study found that FZ in FATP4 gene expression trends are increasing - reducing - increasing. The study found that SMBC and S01 expression is not a significant difference, but the expression of SMBC is higher than the expression of S01.
