In this research,the polymorphisms of cattle NOD2 gene and the relationship between the polymorphisms of cattle NOD2 gene and cattle mastiffs in 304 cattles including Chinese Holstain cattle,Chinese simmental cattle,Tongjiang huangniu,Swamp buffalo and chinese yak were detected by PCR-SSCP test Based on the detection of polymophisms,the effects of different genotypes on milk qulitaty and scs were analyzed.With eight different pairs of primers,expand exon 8,11 and 12.There were two Single Nucleotide Polymorphism(SNP) locis on the extron 12,SNP+144(A→T) on the prime 1, SNP+1594(G→A) on the prime 5.There were two alleles of SNP+144,A and B. Examines three kind of genotypes in the Chinese Holstaine cattle,and Chinese simmental cattle,it's AA,AB and BB.But only two kind of genotypes examined AA and AB in the Tongjiang huangniu,Swamp buffalo and chinese yak,BB genotype not examined.This locis in Chinese Holstaine cattle,and Chinese simmental cattle,Tongjiang huangniu, Swamp buffalo is demonstrated moderate many condition(0.25<PIC<0.50),demonstrates low in the chinese yak condition(PIC<0.25).There were two alleles of SNP+1594,E and F.Examines three kind of genotypes in the Chinese Holstaine cattle,and Chinese simmental cattle,it's EE,EF and FF.but only two kind of genotypes examined EE and EF in the Tongjiang huangniu,Swamp buffalo and chinese yak,FF genotype not examined.This locis in five breeds cattle is demonstrated moderate many condition (0.25<PIC<0.50).The association between the two polymorphism sites residing in twelve extron of NOD2 gene and body somatic cell score(SCS) of Chinese Holstain cattle and Chinese simmental cattle were analyzed GLM model.The BB genotypes of primel had significant effect on the fat of milk trait than AA,AB genotypes(P<0.05).The BB genotypes of prime1 had significant effect on the fat of milk trait than AA,AB genotypes(P<0.05),The BB genotypes of prime1 had very significant effect on protein of milk and SCS than AA, AB genotypes(P<0.01),The BB genotypes of prime1 had significant effect on the product milk of 305 milk and SCS than AA,AB BB genotypes(P<0.05).The FF genotypes of prime5 had very significant effect on the on protein of milk trait than EE,EF genotypes(P<0.01),The FF genotypes of prime5 had very significant effect on the product milk of 305 milk than EEgenotypes(P<0.05),had significant effect than EF(P<0.05).The genotype combines BBFF had very significant higher on SCS than AAEE,AAEF,ABEE, BBEE and the BBEF genotype combines(P<0.01),had significant effect than AAFF and ABFF(P<0.05).The genotype combines BBFF had very significant higher on total protein of milk than AAEE,AAEF,AAFF,ABEE,ABFF,the BBEE genotype combines(P<0.01), had significant higher than than ABEF and BBEF(P<0.05).The genotype combines AAFF had very significant higher on 305 day milk production quantity than AAEE,ABEE, ABEF,ABFF,BBEE,BBEF and the BBFF genotype combines(P<0.01),the genotype combines AAEF had significant higher on 305 day milk production quantity obviousl than the BBFF genotype combine(P<0.05).Chinese Holstain cattle's SCS had significant higher than Chinese simmental cattle(P<0.05).The NOD2 gene can be used to candidate gene for cow mastitis,305 day milk production quantities,the milk total protein.
