| Aconitum carmichaeli Debx. is perennial-herbage plant which belongs to Ranunculaceae. In this examination, Aconitum carmichaeli Debx. seed 's configuration was observed. Discussing characteristic of dormancy and germination, the Aconitum carmichaeli Debx. seed were process in different temperatrue , stratification, gibberellin, potassium nitrate etc. With fresh pollen as material, pollen structure was observed. In different culture medium, the pollen was cultivate, according to pollen germination rate and pollen tube length, and multi- colouration ways, pollen vigor were studied primarily, and the best suitability culture medium was determined. With Aconitum carmichaeli Debx. seedling as material, according to determine fresh weight, dry weight and aconitine content termly, the test studied on seedling growth trends and aconitine trends. The results are as follows:1. Puce, with thecal wing outside, the Aconitum carmichaeli Debx. seed is coniform wiping off the wing, with brown seed capsule. The seed size is 4.48 mm long, 2.60 mm wide, and 0.97 mm deep,and the seed weight is 2.692 gramme /one thousand seeds. It's largest absorption rate was at 2 hours after seed wetting, and 24 hour later, the seed weight did' nt change anymore.2. The treatment of seed soaking with GA3 and stratification could increase the seed germination value and germination rate effectively, and the maximum germination rate was 94.5%. The germination value and germination rate increased with the temperature raising at normal atmospheric temperature (between 10℃to 20℃). The condition at alternate temperature (10℃～20℃) was favourable for seed germination, and the germination rate was higer than at normal atmospheric temperature. KNO3 was not suited using as a priming reagent. The seed vigor from Beichuan County was higer than from Qingchuan and Pingwu County.3. The seedling of Aconitum carmichaeli Debx. grows slowly, and the most plant have three leaves (some have five leaves) during the seed germination to plant death. It's rhizome growth relyed on the leaves, which can carry out photosynthesis, the more leaves, the biger rhizome,and the biggest rhizome diameter was 14.4 mm, and the length of rhizome was 4 cm.4. With seedling growth, the aconitine content was increasing. The aconitine content was 0.56 mg～1.26 mg/g from Beichuan, Qingchuan's was higer, 1.72 mg/g mostly .5. Ellipse as pollen modality, 42.2 urn by length, 22.35 urn by width, the Aconitum carmichaeli Debx. has three germination foramen, and there's many salience on pollen surface. It's easy to see the modality under the oil and glycerin, but hard to know it in vinegar- magenta because of changing shape through drinking water.6. Boron can promote pollen germination even minim content, but there's no much fluence that higher or less the boron content has, and pollen cannot germinate lacking boron. Pollen germination rate and pollen tube length ruduced with cane sugar increasing, and pollen mostly cannot germinate while lacking or arriving at 25% the cane sugar was. Temperature is important, pollen germination rate and pollen tube length in 25℃were higer than in 5℃and in 30℃. The optimum condition of pollen germination was 25℃, 0.8% agar +5% sugar +50mg/L boric acid. |
