To investigate the characteristics of potassium accumulation in AtCIPK23-introduced K326 andYuyan 6, Sand Culture and Water culture, combined with field experiments, were used. The results were as follows:AtCIPK23, a protein kinase, interacting with CBL1/9, is crucial to the response to low-K+ stress in Arabidopsis. In this study, genes of transgenic tobacco AtCIPK23 treated with potassium at different levels was found resistant to low potassium stress. Its growth was less inhibited with more roots, large vegetative organ and normal leaf colour. The result in Hypokalemia treatments showed that the biomass of transgenic tobacco increased by 54.5% compared to wild-type and NC89, the content of leaf potassium in transgenic tobacco was 0.154 and 0.04 times more than in wild-type and NC89 respectively. The study on potassium absorption kinetics indicated that KM of transgenic tobacco dereased by 48.5% and 56.14% compared to wild-type and NC89 respectively, Cmin decreased by 47.37% and 41.18% respectively, and thereas Vmax showed no significant difference. It was postulated that AtCIPK23 is more resistant to low potassium stress.The results by Sand Culture and Water culture and field experiments showed that the total dry weight of No.6 yuyan was higher than that of K326 by 45.14 percent significantly at the low potassium levels, and at high potassium levels , its dry weight was 2.92g higher than K326's largely significantly. No.6 yuyan had 7.693g of root fresh weight, 3.333ml of root volume more than K326 significantly, 480.538μg / (g.FW.h) of root activity with a very significant difference. The results of study on Potassium absorption kinetics showed that Vmax of No.6 yuyan was 3.93 times bigger than that of K326 significantly. It indicated that No.6 yuyan posessed developed root system, thus a high rate of potassium absorption.Under field conditions, No.6 yuyan had a best performance supplied with 18 kilograms of pure potassium from which the yield of 150.3 kilograms and the output value of 1833.7 yuan RMB per mu resulted higher than Zhongyan 100 , in varieties for the best performance, was significantly higher than the control tobacco varieties 100 significantly. The content of leaf potassium in No.6 yuyan increased with the potassium supply, but there was no significant difference between the control and No.6 yuyan.
