Molecular Cloning And Distribution Of Pig Zinc Transporter

In order to study the role of the pig zinc transporter protein in the zinc transportation, this experiment searched pig ESTs and UniGene database according to the sequences of human ,Rattus,Sus scrofa and Bos taurus,.then A pair of cloning primers were designed according to the cloning principle of homologous sequence. and RT-PCR was done.Finally,these 9 zinc transporter genes were cloned.And we studied each gene's distribution in the pig's tissue. The main findings are as follows:1,The cloning and distribution of pig zinc transporter protein ZnT-1 gene slc30A1Cloned gene slc30A1 contains a complete open reading frame (open reading frame, ORF),the sequence length is 1521bp, and ZnT-1 protein is encoded by 506 amino acid residues ,which contains 93 acidic amino acids and 66 basic amino acids,and the molecular weight is 55.13 kDa,the isoelectric point is 6.543.The predicted area of peptide is 1-29aa,and the mature protein ZnT-1 is made up of 477 amino acids.This gene was found in the brain, ovary, uterus, breast, esophagus, duodenum, jejunum, caecum, colon, rectum, heart, liver, lung and muscle tissues.2,The cloning and distribution of pig zinc transporter protein ZnT-2 gene slc30A2Cloned gene slc30A2 contains a complete open reading frame (open reading frame, ORF),the sequence length is 1044bp, and ZnT-2 protein is encoded by 348 amino acid residues ,which contains 50 acidic amino acids and 41 basic amino acids,and the molecular weight is 37.95 kDa,the isoelectric point is 6.703. This gene was found in the ovary, esophagus, appendix, rectum, lung and kidney.3,The cloning and distribution of pig zinc transporter protein ZnT-3 gene slc30A3Cloned gene slc30A3 contains a complete open reading frame (open reading frame, ORF),the sequence length is 1164bp, and ZnT-3 protein is encoded by 388 amino acid residues ,which contains 49 acidic amino acids and 42 basic amino acids,and the molecular weight is 41.64kDa,the isoelectric point is 6.501. This gene was found in the brain, ovary, uterus, breast, esophagus, cecum, colon, rectum, heart, liver, lung and kidney.4,The cloning and distribution of pig zinc transporter protein ZnT-4 gene slc30A4Cloned gene slc30A4 contains a complete open reading frame (open reading frame, ORF),the sequence length is 1287bp, and ZnT-4 protein is encoded by 429 amino acid residues ,which contains 72 acidic amino acids and 54 basic amino acids,and the molecular weight is 47.54kDa,the isoelectric point is 6.917. This gene was found in the ovarian, breast, esophagus, duodenum, appendix, heart and lungs.5,The cloning and distribution of pig zinc transporter protein ZnT-5 gene slc30A5Cloned gene slc30A5 contains a complete open reading frame (open reading frame, ORF),the sequence length is 2301bp, and ZnT-5 protein is encoded by 767 amino acid residues ,which contains 94 acidic amino acids and 94 basic amino acids,and the molecular weight is 84.34kDa,the isoelectric point is 7.415. This gene was found in the brain, ovarian, breast, esophagus, jejunum, caecum, colon, rectum, heart, liver, lung and kidney.6,The cloning and distribution of pig zinc transporter protein ZnT-6 gene slc30A6Cloned gene slc30A6 contains a complete open reading frame (open reading frame, ORF),the sequence length is 1383bp, and ZnT-6 protein is encoded by 461 amino acid residues ,which contains 63 acidic amino acids and 47 basic amino acids,and the molecular weight is 51.04kDa,the isoelectric point is 9.275. This gene was found in the brain, ovarian, breast, esophagus, duodenum, jejunum, caecum, colon, rectum, heart, liver, lung, kidney and muscle.7,The cloning and distribution of pig zinc transporter protein ZnT-7 gene slc30A7Cloned gene slc30A7 contains a complete open reading frame (open reading frame, ORF),the sequence length is 1128bp, and ZnT-7 protein is encoded by 376 amino acid residues ,which contains 56 acidic amino acids and 49 basic amino acids,and the molecular weight is 41.54kDa,the isoelectric point is 6.943. This gene was found in the brain, ovary, uterus, breast, esophagus, duodenum, caecum, colon, rectum, heart, liver, lung, kidney and muscle.8,The cloning and distribution of pig zinc transporter protein ZnT-8 gene slc30A8Cloned gene slc30A8 is a variant, the sequence length is 1333bp, the biological function has not been studied because of mutations. This gene was found in the ileum.9,The cloning and distribution of pig zinc transporter protein ZnT-9 gene slc30A9Cloned gene slc30A9 contains a complete open reading frame (open reading frame, ORF),the sequence length is 1698bp, and ZnT-9 protein is encoded by 566 amino acid residues ,which contains 108 acidic amino acids and 79 basic amino acids,and the molecular weight is 63.22kDa,the isoelectric point is 8.449. This gene was found in the brain, ovary, uterus, breast, esophagus, duodenum, jejunum, ileum, cecum, colon, rectum, heart, liver, spleen, lung, kidney, pancreas and muscle.
