Study On The β-1,3-glucan-binding Protein-Lipoprotein In The Shrimp Fenneropenaeus Chinensis | | Posted on:2009-11-17 | Degree:Master | Type:Thesis | | Country:China | Candidate:X H Li | Full Text:PDF | | GTID:2143360278971085 | Subject:Botany | | Abstract/Summary: | PDF Full Text Request | | TheβGBP-HDL of Chinese shrimp Fenneropenaeus chinensis was first studied in our experiment.And the experiment provides essential essay datas for further study ofβGBP-HDL.In the present study,a fragment of cDNAs ofβ-1,3-glucan-binding protein-Lipoprotein(βGBP-HDL) was cloned from hepatopancreas of Chinese shrimp Fenneropenaeus chinensis.This fragment shared 93%identity withβGBP-HDL of shrimp Litopenaeus vannamei.RT-PCR assays provided evidences that theβGBP-HDL was synthesized in hepatopancreas,hemocyte and gut.TheβGBP-HDL constitutively express at every stage in the shrimp Fenneropenaeus chinensis.Using semi-quantitative RT-PCR,stress protein induction data at different time from the shrimp Fenneropenaeus chinensis during starvation stresses was obtained.A maximum value ofβGBP-HDL mRNA was observed at 2 h after ingestion,then after 24 h a down-regulation with a reduction of 1.7-fold on the mRNA level in comparison to 2 h starvation was detected.This was followed by a small recovery at 48,72 and 120 h;nevertheless.We also found thatβGBP-HDL mRNA level in shrimp of high-fat feeding were higher than that of normai group.This indicates thatβGBP-HDL involved in lipid transportation.The relative expression ofβGBP-HDL after infected by WSSV was studied by real-time PCR.The results were as follows:After infection,the relative expression ofβGBP-HDL was higher than normal shrimps.At hour 6 post-infection,the expression reached the maximum quantity.The study indicatesβGBP-HDL may be involved in the defense response against virus infection. | | Keywords/Search Tags: | shrimp Fenneropenaeus chinensis, β-1,3-glucan-binding protein, molecular cloning, real-time PCR, Whitespot Syndrom Virus(WSSV) | PDF Full Text Request | Related items |
