The tumor is a kind of disease, and the human's life and health are threaten by which seriously. The treatment for cancer such as surgery, chemotherapy, radiotherapy etc, there are some serious damages to body. In recently years, the scholars all over the world have given the enormous attention to the anti-tumor natural medicines which poisonous side effects are small and active functions are widespread. Paclitaxel is one kind of natural medicines which have the anti-tumor activeness. As an anticancer medicine developed in 20 century's end, Paclitaxel has universal applied on the clinical care of cancer diseases, such as the ovarian cancer, the breast cancer, the nonsmall-cellule lung cancer and melanoma, whose rominent curative effect has caused widespread attention. But the apoptosis mechanism of the canine breast tumor cell induced by the Paclitaxel has not a perfect explanation at present, this experiment has conducted a series of studies on this question.This experiment used breast tumor cells in vitro as the materials that was treated by Paclitaxel with 0 nmol/L, 4 nmol/L, 8 nmol/L and 16 nmol/L respectively. Then observed the apoptosis changes using the BrdU fluorescence marking method, detected the inhibition ratio using the MTT method, detected the activity of Caspase-3 and Caspase-6, the concentration of Ca2+, the contents of Cyt c using the kit chromatometry method, detected the cell factors level using the radioimmunity method at 24h, 48h and 72h separately post-treatment with Paclitaxel. The results showed as follows:The morphology results showed that the Paclitaxel could inhibit the cell multiplication by BrdU fluorescence marking method. The inhibition ratio revealed that the canine breast tumor cell is supersensitibe to Paclitaxel, and the sensitibity increased along with times going and dose increasing.Compared with control cells, the contents of G-6-PDH has decreased, the results demonstrated that Paclitaxel inhibited the cell growth through destructing the cell membrane integrity and affecting DNA and RNA synthesis.The activities of Caspase-3 and Caspase-6, the concentration of Ca2+ and the contents of Cyt c in the canine breast tumor also have obvious change, the detections were demonstrated that Pacliaxel can stimulate the apoptosis signal pathways to induce the canine breast tumor cell apoptosis. It was found for the first time that Paclitaxel can induce the canine breast tumor cell to poroduce IL-1, IL-2, IL-6, IL-8 and TNF-αin supernate. This may be the cause and mechanism of inhibition of cell multiplication occurred possibly. |
