The activities of antioxidant ingredient extracted from Garcinia mangostana's pericarp measured with FRAP model, reducibility measuring and DPPH free radical's scavenging efficiency tests in this paper. Results showed that the best solvent extracting antioxidant ingredient from Garcinia mangostana's pericarp is 95% ethanol. According to orthogonal experiment and single factor comparison experiment, the optimized process is as follows: soaking time was 3 hours, pH value was 7, solid-liquid ratio was 1:20, extracting temperature was 70℃. Then the heat stability of the 95% ethanol extracts was studied. It showed that, the antioxidant activity of 95% ethanol extracts became stronger and stronger with the increasing of the temperature from 0°C to 60°C, while it decreased with the prolonging of the preserving time.The solvent classifications method was used to separate the 95% ethanol extracts. And benzin petroleun extracts, ethyl acetate extracts, n-butanol extracts, aqueous extracts were collected. Then their antioxidant activities were assayed by external antioxidant mode. Besides, the content of flavonoids and polyphenols were determined. The result showed that: Antioxidant activities of n-butanol extracts is stronger and the content of flavonoids and polyphenols were higher than the other parts.The n-butanol extracts were purified with adsorb resin D101. and then the content of flavonoids and polyphenols were determined. After purified the n-butanol extracts, it showed that the antioxidant activities and the content of flavonoids and polyphenols were remarkably enhanced.The n-butanol extract was purified with adsorb resin D101.The antioxidant activity of the purification was measured with the restraining mice brain and liver tissue homogenates lipid peroxidantion method and other antioxidant models. It showed that: the antioxidant activity of the purification was strong in these tests.The dried pericarp of G. mangostana was extracted by 95% ethanol for 5h, twice. After filteration and evaporation of the solvent under reducedpressure, the combined crude ethanolic extract was suspended in H2O to produce an aqueous solution, then partitioned in turn with benzin petroleum, EtOAc and n-BuOH. The different extracts were subjected to chromatography over a silica gel column, prep TLC. crystallization et al, to give one pure compound. Structure of the compound have been established and identified on the basis of chemical and spectroscopic evidence (1H-NMR, 13C-NMR, 2DNMR. MS). It isα-mangostin.
