In order to investigate the expression changes of antimicrobial peptide of day and night in chicken′s intestine, we used immunohistochemistry to study the dynamic changes of expression of antimicrobial peptide in two-week-age roman chicken′s intestine. Experimental design and results are as follows:after the free food intake in a week, 1-day-old chicks were taken strict light′s control, Specific approach is to give light at 8:00 am and remove light at 8:00 pm in order to ensure the formation of the normal biological clock . We drown the materials when the chickens were Two-week-old. during 24 hours of a day ,we divided it into 8 time points that is 8:00,11:00,14:00,17:00,20:00,23:00,2:00 in the next day,5:00 in the next day,and drown the materials at the 8 time points,and drown the materials at 8:00 of next day repeatedly, and needed 9 times Totally.we took five chickens randomly, we Dissected and took out of their intestinal sections,and put their intestinal sections into a bottle filled with formalin for fixation and Preservation for uesing later, After the chickens were anesthetized with ether. We made paraffin sections of Intestinal tissue by conventional methods ,and detecte expression of antimicrobial peptides in Intestinal sections by immunohistochemical method, and determined the area of the positive cells in the unit area in one field of vision with micro-Leica image processing system and QWin software, and took this as quantitative criterion of expression of antimicrobial peptides.The results showed that: diurnal variation of expression of antimicrobial peptide in Chicken′s intestine shows certain regularity Generally. From the morning to the next morning the expression is at first increased and then decreased again and has a downward trend at last. The expression of antimicrobial peptides is in the middle level at 8:00 am. After stimulation of light during the day, the level of expression of intestinal antimicrobial peptide tends to decline in general. The expression of intestinal antimicrobial peptides is at a lower level at 14:00 and 17:00.The level of expression of intestinal antimicrobial peptide begans to rebound at 20:00,and its level of expression close to the levels at 8:00. Its expression continue to increase at 23:00. expression of the antimicrobial peptide is at a higher level at 2:00 am and 5:00 am,and bring back to The initial level at 8:00am in next day. In addition, the levels of expression of antimicrobial peptides in protoduodenum,duodenal disruption and metaduodenum at the same time point have a little difference,but the trends of the expression of antimicrobial peptide in protoduodenum,duodenal disruption and metaduodenum is consistent at different time points. The trends of expression of antimicrobial peptides in jejunum, yolk stalk and rectum is different with the general trend at different time points. This difference embodies at the several time points of the beginning mainly,theirs trends are different with the trends of the beginning in other intestinal sections,and show upward trend slightly at the first. However, the expression of antimicrobial peptide in this three intestinal sections are still at a relatively lower level during the day. the expression levels of antimicrobial peptides achieve the highest level at 2:00 and 5:00 during a day. This is consistent with the trend of the expression of antimicrobial peptide in other intestinal sections. The trends of expression of antimicrobial peptides in ileum and cecum are Consistent with that in duodenum. Overall,the average level of expression of antimicrobial peptides in ileum is higher than that in other intestinal sections, and its range of diurnal variation is also relatively large. the average level of expression of antimicrobial peptides in rectum is higher than that in other intestinal sections, and its range of diurnal variation is Small.It shows that diurnal variation of expression of antimicrobial peptide in Chicken′s intestine shows certain regularity, through the role of Reset of the clock,the clock keep chicken expression of antimicrobial peptide in Chicken′s intestine in a state of relative stability during a whole day.this is of great significance to play full role of antimicrobial peptides in immune defense.
