microRNA(miRNAs) are 22nt non-coding RNA that have been shown to inner regulate gene expression in variety of physiological function for organism. The expression analysis, as with functional exploration had been carried out in model organisms, therefore, miRNA resreaches have been a hotspot in biology at present. They don't only express in embryo development, but also have different expression in adult.As one ofβ-adrenergic agonists, clenbuterol is harmful to human being health after helping to improve procine meat quality traits. Some genes can increase skeletal muscle mass and decreases body fat in livestock treated by clenbuterol. We analyzed microRNAs differential expression of muscle tissues of Chinese miniature Swines with/without clenbuterol and built relationship with target genes.We detected 13 microRNAs (miR-1,miR-133,miR-206,miR-29a,miR-29b,miR-181a,miR-193,miR-195,miR-196a,let-7a,let-7b,miR-26a,miR-30d) from muscle tissue of Chinese miniature Swines by northern blot. miR-133,miR-206,miR-1 show the bright band, the others show the weak band such as miR-181a,miR-193,miR-195. Then, we detected the expression of microRNA by real-time PCR. The results were as follows:1. The expression of miR-133,miR-206, miR-1 was higher than other microRNAs. But they had no different between with or without clenbuterol.2. let-7a and miR-26a had no different between with or without clenbuterol, but their expression were decline as long as grown.3. The expression of miR-181a,miR-193,miR-195,miR-196a,miR-30d,let-7b had no significant difference.4. The expression of miR-29a,miR-29b had significant difference, they were opposite.5. Predicted the target genes of microRNAs:1) The microRNAs which have no difference between with/without clenbuterol also had target on these genes.2) miR-29a,miR-29b had target genes such as Integrin-linked kinase, Elongation factor 1 alpha, MHC class I antigen, SWI/SNF complex, HnRNP F protein. Then we analyzed the influence of microRNAs and target genes on the phenotype of muscle with clenbuterol.
