In this study,selected animals and people in Jilin Province for the study of the prevalence of zoonotic HEV investigation analysis.To obtain the serological evidence of hepatitis E virus(HEV) infection in different animal species and different groups of people, we collected 1208 sera from big pigs(age greater than 3-month-old),1303sera from piglets (age less than 3-month-old),890sera from Cattle,729 sera from sheep,643 sera from general crowd,426 sera from pigs-occupational crowd and 296sera from non-pig occupational crowd and detected the antibody and antigen to HEV by enzyme-linked immunosorbent assay(ELISA) with the kit(HEV Ag kit and animals used HEV Ab kit,people used HEV IgG,Ig M kit) from Beijing Wantai biological medicine products company.Survey results show that:①HEV infection is common of pig,and the infection rate of big pigs were significantly higher than piglets,pig has the highest level of HEV antibodies,pig is more likely vulnerable to HEV than other populations.②Pigs-occupational crowd infected HEV ratio also very impressive,the prevalence rate of pigs-occupational crowd was higher than general crowd,men higher than women.③22 sera from pigs,18sera from cattle,10sera from sheep,2 sera from crowd were also detected to be HEV Ag positive samples and 4 sera of crowd HEV IgM positive samples in the JiLin provinces.④In the three areas crowd in Jilin Province,HEV infection is that Songyuan region(saline area) the highest, Changchun region(plains region) followed and Jilin region(Mid-levels district) the lowest.Stool samples Collected from three regions of Jilin,HEV RNA was detected by RT-PCR with partly positive serum and stool samples.The positive PCR products were cloned and sequenced.The sequences were compared with the reported HEV isolates.The sequence analysis showed that the homology of the 7 HEV RNA positive clones was 86.2%~98.6%,that is HEVⅣ.They belong to the same branch through the analysis of phylogeny. |
