This thesis main study is low temperature keep-alive of turbots(Scophthalmus maximus).Through improving and developing the existing keep-alive methods,it can have longer keep-alive time and higher survival rate.The data were analyzed by principle component analysis method,multivariate analysis method,liquid chromatography on turbot muscle ATP contents,the data is expressed by average value and standard deviation,analyzed by significant differences.The main research reports are:Using different concentrations of eugenol anesthesia turbots at 9~10℃. According to characteristic behaviors of turbot anesthesia,it can distinguish anesthesia stage.According to characteristic behaviors of anesthesia turbots in clean seawater,it can distinguish resuscitation stage.Through comparing anesthesia time of different concentrations of eugenol,resuscitation time and survival rate,it can summarize the effective concentration on turbots is 20~40mg/L.Different voltages and different time electric shock on turbots in seawater at 9~10℃.Respectively do the experiment of different voltages on turbot electric shock effects,different time on turbot electric shock effects,and at the same shock condition, electric shock effects on different turbot size.Through comparing all kinds of shock conditions,recovery time and survival rate in clean seawater,it can summarize the results.The results are,as the shock voltage is higher,the electric shock effects are more significant.As the shock time is longer,the electric shock effects are more significant.At the same shock condition,turbots of bigger size are hard to anesthesia, but the recovery speed of bigger size turbots is slower than small size. It is the first time to explore to keep turbots alive after anesthesia by eugenol and electric shock at 10℃.Through comparing different characteristic behaviors between keep-alive with and without water,and the survival rate during keep-alive and recovery process,it can summarize the results.The results show that turbots after anesthesia by eugenol and electric shock,can increase keep-alive without water time and survival rate.The results propose the flounder fish after anesthesia by eugenol keep alive without water,have high practical feasibility.Successful to attempt to keep turbots alive at 3℃,the survival rate is 95%for 60 hours.Examination turbots blood hematological and biochemical indices during keeping alive at 3℃,the results are obtained by horizontal and vertical comparison. The results show that during keeping alive,the MCV get bigger obviously,the reason maybe is turbots try to adapt to hypoxia environment.During turbots keeping alive, blood indices(BUN,CR,UA) have a rising trend as time increasing,and blood indices(GLU,CHE) have a downward trend.The blood indices(BUN,CR,UA,GLU) of keep-alive with water are obviously lower than keep-alive without water. Through the analysis on hematological and biochemical indices,the main reason of turbots death is that the metabolites can not excrete effectively,and the excessive content of Ammonia Nitrogen directly caused turbots dead.
