The germination rate of Anoectochilus roxburghii seeds under different conditions and the effects of four factors(minemun culture medium, 6-BA,ZT,and NAA) on protocorm multiplication under orthogonal design were studied,as well as the desiccation tolerance of protocom was discussed.The results are as follows:1.Pollination type was an important factor on A.roxburghii seeds' non-symbiotic germination,germination ratio of the xenogamy seeds reached 78.53%,and the germination ratio of the geitonogamy seeds reached 69.62%,while the germination ratio of self-pollination was only 39.87%;the most suitable maturity of capsule appeared when it was 150 days old,the germination rate of such seeds could reach 78.59%; refrigerated storage affected the vitality of the seeds:the germination rate of the seeds was decreased with the increase of seeds' refrigerated storage time;compared with the seeds treated by sodium hypochlorite solution there was no obvious difference on germination rate;2.Variance analysis showed that NAA has a strong effect on the multiplication of protocorm and that the optimal medium for protocorm multiplication is 1/2MS+ZT0.5mg/L+NAA1.0mg/L.3.The content of soluable protein and heat-stable protein of protocorm took on the change of "M" type.After SDS-PAGE,compared with the control,a novel protein with about 28.6kDa was expressed when the protocorm was treated in 30%PEG-6000 for 2ds,3ds,4ds,5ds and in 40%PEG-6000 for 1ds,2ds,3ds,4ds,5ds.The novel protein was heat-stable. |
