Macrophages are the basis of animal immune response play an important role in host defense in phagocytosis, cytotoxicity, anti-presenting, cytokines secretion, regulating internal environment and antitumor effects. In previous study, we found that Polysavone increased the immunity in broilers and pigs , promote the proliferation of lymphocytes. To further investigate its effects on immune system and its mechanism in mouse macrophages, Polysavone and its active ingredients (polysaccharide, flavonoids, saponins) with different dose were used to culture RAW264.7 cells, and the mRNA expression of different kinds of mouse beta defensins (mBDs) were detected.Exp. One Effects of Polysavone on expression of mouse beta defensins in RAW264.7 cells To investigate the mechanism involved in that Polysavone improve the immunity of animal body.The mouse macrophage line RAW264.7 cells were stimulated by LPS at 0,6,9,12,18,24 h respectively,and the mRNA expression of mBDs was detected. Polysavone with different dose (0.2,0.4,0.6,1.0mg/ml) were used to culture RAW264.7 cells, and the mRNA expression of different kinds of mBDs (mBD1~3) were detected. The results showed that the mBD2 and mBD3 mRNA expression could be improved by LPS, and it is related to the culturing time , but there were no effects on the mRNA expression of mBD1.The medium containing Polysavone could improve the mRNA expression of mBDs, especially the treatment with 0.2mg/ml Polysavone.Exp. Two Effects of polysaccharide, flavonoids and saponins of Polysavone on expression of mouse beta defensins in RAW264.7 cellsTo investigate the effect of polysaccharide, flavonoids and saponins of Polysavone on the expression of mouse beta-defensins in RAW264.7 cells. RAW264.7 cells were cultured with Polysavone polysaccharide at 30,60,90,150μg/ml, Polysavone flavonoids at 10,20,30,50μg/ml, Polysavone saponins at 10,20,30,50μg/ml respectively. The mRNA expression of mBDs (mBD1~3) were detected. The results showed that the mBD2 and mBD3 mRNA expression could be improved by Polysavone polysaccharide, especially the treatment with 30μg/ml, but there were no effects on the mRNA expression of mBD1. The mBD1 mRNA expression could be improved by Polysavone flavonoids, especially the treatment with 10μg/ml, the mBD2 and mBD3 mRNA expression could be inhibited. The mBD1 mRNA expression could be improved by Polysavone saponins, the mBD3 mRNA expression could be inhibited, but there were no effects on the mRNA expression of mBD2. The Polysavone polysaccharide may enhance the function of adaptive immunity. The flavonoids and saponins of Polysavone may enhance the function of innate immunity.Conclusion Polysavone could improve the mRNA expression of mBDs, especially the treatment with 0.2mg/ml Polysavone. The Polysavone polysaccharide may enhance the function of adaptive immunity. The flavonoids and saponins of Polysavone may enhance the function of innate immunity.
