| Kenaf is a ex-situation reproduction and span-year plant.It is the most important topic to maitain the genetic integrity of germplasm resources during kenaf seed conversation.Hence, it has important realistic significance to research physiological, biochemical mechanism and genetic integrity of kenaf seeds during aging.In this study,15 kenaf cultivars were studied.It showed the effects on kenaf seed viability, physiological and biochemical characteristics, heat-stable protein and DNA genetic integrity during difference aging levels,and ascertained the relationship among kenaf seed aging and seed viability, lipid peroxidation protective enzyme systems, heat-stable protein;and understood aging-resistant cultivars. The results are as follows:1. 15 accessions of kenaf cultivars (Hibiscus cannabinusL.) were studied by artificial acceleration aging method, the results demonstrated:seed gemination and vigor index of these materials reduced gradually under the artifical conditions; they decreased as a uneven speed with the extent time in the process of aging;the 15 variaties of kenaf were ordered in sequence from strong to weak by Membership Function as follows: Liao 55 > C-2> Yuehong1hao > SD124 > Xinhong95 > Taihong763>KB11> Qingpi1 >83-20>SF192>V379>Hongyin135>KB2>Fonghong991>Tainong1. The germination potential,germination percentage,germination index, vigor index, simple vigor index and seedling dry weight wer negatively correlated with extent time of artificial aging treatment.2. Firstly,POD and CAT activity in kenaf seed increased during aging process. And following they decreased and up to the lowest level;in contrary, MDA content decreased at first, then increased, finally reached to the highest level.By correlation analysis, POD and CAT activity had positive correlation and enhanced anti-oxidation by mutual coordination, improving anti-aging ability of.seed POD activity and MDA content showed negative correlation,of which Fuhong991,Hongyin 135 had significantly negative correlation, correlation coefficients were -0.8125, -0.8276;CAT activity and MDA content showed negative correlation, Taiong1hao,Hongyin135,KB2 significantly negative correlation,correlation coefficients -0.8434, -0.7236, -0.8243.3. 10 accessions of kenaf cultivars were studied on heat-stable proteins The results indicated that,the content of heat-stable protein gradually increased with aging treatment deepening,SDS-PAGE eletrophoresis showed that a 62.8kD heat-stable protein appeared only in liao 55 in 140h aging treatment,there were no difference in others.Liao 55 was the most anti-aging variety during aging treatment in 10 kenaf varieties,these results suggested that the the special heat-stable proteins might be related to aging tolerance.4. 3 accessions of kenaf cultivars had beed detected by RAPD markers, after different aging treatment,the allele frequencies of Tainong No.1 and 83-20 were 100%,no differences detected; Liao55 100.85%, there were no significant difference by t-testIt showed that aging treatment had no effect on genetic integrity of often cross-pollinated crop of kenaf. |
