Ips typographus,one of the most damaging bark beetles on the Eurasia,mainly distributed in Heilongjiang,Jilin,Qinghai and other areas in China.Recent years,outbreaks of I. Pypographus have brought the coniferous forest lots of damages in easten Jiling and Heilongjiang Province.All the researches on I.Pypographus in Northeast China were focused on the biology and chemical ecology,and the results of those researches were disaeeord.As evidences showen above provide a reason to suspect that there may be different population in Northeast China.I.typographus popultions were analysed by mitochondrial DNA 12S rRNA,COâ… and COâ…¡genes of different geographic samples.First extracted genome DNA,then amplified these three gene fragments of mitochondrial DNA,later mensurated those sequences.Nucleotides composition,ratio of transition and transversion,genetic distance,considerable variation of this segment were analyzed.Based on these three gene groups,the molecular phylogeny trees were built by using Neighbor-Joining(NJ),Minimum Evolution(ME),Maximum Parsimony (MP) and Unweighted Pair Group Method with Arithmatic Mean(UPGMA) methods.The results indicated that there was no alien population and population differentiation of I. pytographus in northeast China.This conclusion provide a foundation molecular systematics research for the unified biology and chemical ecology research approach to protect and control the damage of Chinese geographical population of I.typographus.This study also analyzed the accuracy of the sequencing results combining with other relevant sequence data of I.typograthus in Genbank to eliminate the results ofpseudogene.And In this paper,the gene molecular phylogenetic tree also constructed based on all the mtDNA-COâ… gene data in Genbank and this study,the result showen that population differentiation was coincident with the geography differentiation.As showen in the molecular phylogenetie tree,I. typograthus divided into two geographic groups:Asian group and European group and the Asian group included Chinese population and the population of Russia.
