FMDV is the causative agent of FMD,a highly contagious disease of cloven-hoofed animals,in addition countries and the people's living by severely affected. FMD in many countries severely limits its trade in animals and animals products due to international regulations designed to limit spread of the disease. And it was paid more attention by OIE. The VP1 gene is the main antigenic epitopes of FMDV which plays very important roles in inducing neutralization antibodies and make protective function.In this study, got the VP1 product from recombinant plasmid of pGEM-KZ-VP1, then ligated the VP1 gene of FMDV of Asia I and the pET-28a expression vector, then transformed into BL21 cells and induced the recombinant plasmid with different concentration of IPTG and different time to express the VP1 protein. The results of SDS-PAGE and Western blot demonstrated that the expressed VP1 protein was existed in the form of inclusion body.Analyzing the effect of different concentration of IPTG and different time on expression product,and confirmed the best induced concentration of IPTG and time. Experimental results indicated that when the concentration of IPTG increased, the products have not increased, and with the time past the products increased. The ultima concentration of IPTG was 3mmol/L and time was 9h, which is better condition when induced VP1 protein.
