In this paper, 22 varieties of flue-cured tobacco were used as materials to investigate the genetic diversity and genetic relationships with RAPD and Morphological Markers. The major results were as follows:1. 38 DNA bands were amplified with 7 RAPD primers, among which, 33 bands (86.8%) were polymorphic.2. Using software Bio ID++,UPGMA cluster analysis was carried out and a dendrogram was constructed. The 22 varieties could be clustered into three groups at the level of D=0.30, and six groups at the level of D=0.23. The six groups included Yunyan and Tezi401 series, Majority varieties from American, 9804,Yanyan97 and Coker176, CF965 and K07, Hongda, C2.3. A dendrogram was constructed based on the Morphological data and the integrated data, respectively. According to the results, the varieties could be clustered into two groups at the level of D=11, and four groups (â…¡1,â…¡2,â…¡3,â…¡4) at the level of D=7.86. Each group of the four groups was clustered into two small groups.â…¡1 group included Yunyan series,CF965 and Miyejian.â…¡2 group inclued the American species. But some American species as well as CB-1,F1-35,F1-38,Hongda and Yanyan97 were inâ…¡3 group, and Tezi401 and its progenies was inâ…¡4 group .4. The correlation coefficient was 0.20 between RADP genetic similarity matrix and the morphologic genetic distance matrix, and there was a significant difference. The result showed that the molecular marker polymorphism basically reflected the genetic variation of tobacco. The results of two types of cluster analyses indicated the geographical origin and genealogical origin of the varieties characteristics. Overall, the genetic diversity analysis could be used to breed and modify varieties, and improve breeding efficiency.
