The animal organism's mucous membrane which was the first gateway of pathogen's invasion was the biggest barrier to external environment.Mucosal Immune System(M-IS) refers to the circumference lymphoid organ's mucous membrane part,including the main antigen contact part and the primary reply induction spot as well as some big superficial region(Digestive tract,Respiratory tract,Genitourinary tract,exocrine gland and so on).MIS is the most important means which organism could resistance infection.The Secretion IgA,which may not only counteract,repel pathogen,shield the mucous membrane from the bacterium and the virus,but also promote the function of natural antibacterial factor in exocrine fluid as well as adjust the mucous Mueosal Immune,is the main effective factor of MIS.As a result of its special position,The Mucosal Immune is playing the influential role in the interaction of organism and the external environment.And the immunity active ingredient in the MIS brings to people's attention more and more.The paper want to reflect approximate the distributed condition of mucosal IgA in the digestive tract,the respiratory tract,the genitourinary tract as detecting IgA content in the oral cavity,stomach(rumen,reticulum, petal stomach,abomasum),duodenum,jejunum,ileum,cecum,colon,rectum,nasal vavity,bronchial tube, lung,urinary bladder,urine,tear fluid,bile,tonsil of beef cattle by double antibody sandwich-Enzyme linked immunosorbent assay(DAS-ELISA).Three parts are following:1.Preface.The MIS and its function,response process of Mucosal Immune,cellular immunity and humoral immunity of the Mucosal Immune are outlined.The new content is increased in the traditional immunology by the MIS research.And the new mentality and method are provided to some disease's pathogenesis mechanisms,the relation between pathogenesis and hosts,diagnosis of disease,new research in vaccine and prevention preparation.2.Straight matter.In the study,IgA was separated and purified directly from milk serum which was centrifuged from cow's colostrum as 10000r/min by Sephadex-G200 gel filtration and DEAE-52.Then it was identified by SDS-PAGE.Then the antibody of rabbit anti-cattle IgA was prepared and labeled with HRP. Meanwhile the antibody of mouse anti-cattle IgA was prepared.At last the gathering various mucous membranes spots(each spot 30 samples) were detected by DAS-ELISA which established in this paper.The content of IgA can calculate from Regression Equation of DAS-ELISA OD value with Content of IgA,and then analyze and discuss the results.3.Conclusions.The technique of DAS-ELISA which could detect the mucosal IgA of beef cattle was established successfully.Then the samples were detected by the technique.And the results are following:the IgA could be secreted in Digesting system,Respiratory system,Urinary system,tear fluid,bile,tonsil.The content of IgA is more in the following parts:rumen,bile,jejunum,rectum,reticulum,and tonsil.And the less ones are:duodenum,abomasum and cecum.
