Foxtail mille(tSetaria italica(L.)Beauv.) is a kind of minor crop which is nutritious, edible, and health with high feeding value. In this study, crude protein, crude fat and vitamin E and other components of 400 accessions primary core-collection of foxtail millet were determined and evaluated. The genetic diversity of primary core-collections was studied in aspects of origins, quality traits and molecular tools.During the process of quality determination and evaluation, the high performance liquid chromatography (HPLC) technology applicable for determining vitamin E in foxtail millet was established, and the classification criteria for determining vitamin E was also established. The results showed that the average content of protein was 15.73%, ranging from 10.86% to 20.87%; the average content of fat was 3. 87%,ranging from 2.38% to 4.92%; the average content of vitamin E,α-T, (β+γ)-T andδ-T were 52.00μg/g, 14.58μg/g,37.31μg/g and 0.52μg/g respectively. 20 accessions of foxtail millets with high contents of crude protein, crude fat and vitamin E were selected.Correlative analysis of the origins, grain color and the main quality traits showed that, the crude protein and the crude fat content of millet from northwest inland were higher than that of other ecological zones; millet from south of Huai River region contained the highest content of vitamin E and (β+γ)-T, and resources from Northeast plains have mostα-T. The crude protein and fat content of red and black grain colors of millets were higher than that of other colors. Black and brown grains contained the highest vitamin E andα-T. Correlative main quality traits analysis of millet indicated that crude protein, crude fat, vitamin E,α-T and (β+γ)-T content were significantly positive correlated with each other.In SSR analysis, a total of 215 alleles were detected using 20 SSR markers, the number of alleles per locus ranged from 3 to 18, with average of 10.75. The mean of polymorphism information content(PIC)was 0.774. PIC of p88 was 0.921 which was the highest, while that of p85 was 0.513 which was the lowest. Shannon-weaver index ranged from 0.959 to 1.872, with average of 1.57. Shannon-weaver index of northwest inland accessions was the highest, while South of Huai river was the lowest.In cluster analysis, 400 accessions primary core-collection of foxtail millet were divided into 7 clusters, each cluster had its own characteristic in quality traits. The average of crude fat, vitamin E,α-T and (β+γ)-T contents of clusterⅤwere the highest,The average crude protein content of clusterⅥwas the highest; while crude protein , crude fat, vitamin E,α-T and (β+γ)-T contents of clusterⅡwere the lowest. |
