Sperm acrosome reaction (AR) is a prerequisite for animal fertilization, the study on sperm acrosome reaction is important for both animal breeding theory and the practice.In this study, the Blue Fox was the experimental animal, and the effect that the calcium ionophore A23187 induced the Blue Fox sperm acrosome reaction was stduied.The appropriate concentration and optimal action time of calcium ionophore A23187 inducing acrosome reaction were screened in this study.The impact of DMSO (solvent of A23187) on the acrosome reaction rate and the survival rate of Blue Fox sperm capacitated or uncapacitated were also discussed.According to the report and pre-experimental results, nine different concentration of A23187 and six different concentration of DMSO were screened in this study .At the same time, the influence of acrosome reaction of the Blue Fox sperm induced by A23187 while the existence of BSA was discussed.The acrosome state, survival rate of blue fox sperm was examined by Coomassie blue staining, Eosin-nigrosine staining, respectively.The results were as follows:1. Within two hours, the acrosome reaction and survival rate of uncapacitated blue fox sperm was not significantly influenced by the treatment of DMSO (0.1%, 0.2% and 0.5%). With the concentration of DMSO increased, the survival rate of blue fox sperm decreased significantly (P<0.05).2. The acrosome reaction rate of capacitated Blue Fox sperm was promoted by DMSO (0.1%, 0.2% and 0.5%). The survival rate of blue fox sperm decreased significantly (P<0.05), when the concentration of DMSO was high.3. The concentration of DMSO should be controlled below 0.5% in Blue Fox sperm capacitation and acrosome reaction.4. A23187 can improve the acrosome reaction of capacitated Blue Fox sperm.For Blue Fox sperm capacitation and acrosome reaction, the most appropriate concentration of A23187 to the blue fox is 5μmol/L, and the most appropriate time is 7 minutes.5. The acrosome reaction rate of uncapacitated Blue Fox sperm was not significantly influenced by the treatment of A23187 (P> 0.05).6. The acrosome reaction of the Blue Fox sperm induced by A23187 was not significantly influenced by the treatment of BSA.
