Abstract/Summary:
This Paper is about the plantlet regeneration process of the main effect factors of Phlox Paniculata and Phlox Subulata,which contains callus and adventitious buds induced,proliferation subculture,rooting,transplanting et al,the antibiotic sensitivities were conducted,and receptor system for the gene transformation of different explants of two species Phlox were established.The result of this research established a foundation for application genetic engineering technology to improve the drought stress tolerance of groundcover plants, and cultivate new varieties.And when we obtain the resistent groundcover plants,that have the important effects on development about water saving and remission using water pressure for freshwater resources shortage's urban,and providing new greening materials for the dryness section of barren.In Phlox Paniculata,with axillary as explants,basal MS media supplemented with 2.0mg/L 6-benzoyladenine and 0.1mg/L a-Naphthalene acetic acid was the optimum media with 91%budding;MS plus 1.0 mg/L 6-Benzoyladenine and 0.1mg/L a-Naphthalene acetic acid was highest shoot proliferation efficiency of 3.1.The leaves of marinating 5 minutes with 5000mg/L vitaminC were used to develop the regeneration system by adventitious buds.With in vitro leaf as explants,the regeneration rate of 86%was achieved in MS media supplemented with 6-Benzoyladenine3.0 mg/L and a-Naphthalene acetic acid 0.1mg/L,and multiplication quotiety achieved 3.7.The 1/2MS media supplemented with a-Naphthalene acetic acid 0.1mg/L and active carbon0.3%and 1/2MS medium supplemented with a-Naphthalene acetic acid 0.2mg/L and active carbon 0.1%was as optimum shoot rooting media with 80%rooting. The rate of surviving achieved 96%.In Phlox Subulata,axillaries as explants,MS media supplemented with 6-benzoyladenine0.4mg/L,Indole-3-butyric acid 0.04mg/L and Gibberellins1.0 mg/L and MS media supplemented with 6-benzoyladenine0.4 mg/L and a-Naphthalene acetic acid 1.5mg/L was as the optimum media with 100%budding;MS media supplemented with 6-benzoyladenine0.4mg/L,Indole-3-butyric acid 0.04mg/L and Gibberellins1.0 mg/L was with highest rate of callus induction of 96%,callus multiplication of 5.6,adventitious buds differentiation of 100%,adventitious buds reproduction of 11.6.The MS media supplemented with 6-benzoyladenine 0.4mg/L and Indole-3-butyric acid 0.04mg/L was the strong seedling culture mediums.The 1/2MS medium supplemented with a-Naphthalene acetic acid 0.2mg/L,Indole-3-acetic acid 0.1 mg/L and 6- benzoyladenine 0.02mg/L as optimum shoot rooting media with 88%rooting.The rate of surviving achieved 85.7%.The experiment about antibiotic sensitivities were conducted,and receptor regeneration system for the gene transformation of two Phlox were established.The results showed that in Phlox Paniculata,the optimum selectivity antibiotic was Kanamycin,the optimum concentration was 200mg/L,the optimum bacteriostasis' was Cefotaxime,the optimum concentration was 50mg/L,which the 50mg/L Cefotaxime controlled the growth of agrobacterium,and the rate of regeneration was 55%;in Phlox Subulata,the optimum selectivity antibiotic was Kanamycin,the optimum concentration was 200mg/L,the optimum bacteriostasis' was Carbenicillin,the optimum concentration was 100mg/L,the 100mg/L Carbenicillin controlled the growth of agrobacterium,and the rate of regeneration was 65%.
