This Paper studied the polysaccharides of Allium Mongolia Regel testing methods, the water extraction method, the protein removal method and immune function were studied, the results were summarized as follows:(1)The optimal condition for the extraction of polysaccharide by hot water was established.The effects of extraction time, extraction temperature and material to water ratio on the yield of polysaccharide were investigated. The optimal parameters were: extraction time 6h, extraction temperature 90℃and material to water ratio 1:20, the yield of onion polysaccharide reached 2.56% under the condition established.(2)Comparing the Sevage method and Trichloroacetic acid method on the protein, the optimization Sevage condition to extract protein from Allium Mongolia Regel was determined. The quantity addition of Sevage, the volume of CHCl3 and n-butanol, frequency of deproteinization and influence of deproteinization were studied. The optimization was found. The quantity of Sevage was 1/4 volume of Allium Mongolia Regel polycose solution; the rate of CHCl3 and n-butanol was 3:1; the deproteinization time was 30min. The deproteinization rate was 43.82%, and the lackage rate of polucose was 4.67%.(3)The immunity influence of Allium Mongolia Regel polycose was studied. The optimization of Allium Mongolia Regel polycose was 75mg/kg·BW·d. The immune organ exponential influence of Allium Mongolia Regel polycose was detected. There was no significant deviation between blank control group and test group in 7d and 14d respective in spleen index(P > 0.05). On the thymus index, there was no significant deviation between blank control group and test group on 7d(P > 0.05), but on the 14d the difference was significant(P < 0.05). The immune function influence of Allium Mongolia Regel polycose was also detected. The activity difference of ACP in serum among the blank group, positive group and test was no significant on the 7d and 14d respctive(P > 0.05). But in the live, the test group was significant higher than blank group on the 14d(P < 0.05). The macrophage function influence of Allium Mongolia Regel polycose was detected. There was no significant difference in carbon clearance index on the 7d and 14d(P > 0.05). But in the index of phagocytize, the test group was higher than blank group on the 7d and 14d(P < 0.05).The lisozyma content influence of Allium Mongolia Regel polycose was tested. The content of lisozyma in test group was higher than the blank group on the 7d and 14d7(P < 0.05). But there was no significant difference between test group and positive group (P > 0.05). The IgG and IL-2 were detected in the specific immunity function influence of Allium Mongolia Regel polycose. The content of serum IgG and IL-2 in test group was extremely higher than blank group(P < 0.05)respective. But there was no significant difference between test group and positive group(P > 0.05).Three main indexes of immunological regulation were no-special immunity, cell immunity and humoral immunity. The stand which judged what could be immunological regulation was that two of the indexes of immunological regulation were positive. The three indexes were positive in this test. So the immunological regulationof Allium Mongolia Regel polycose was obviously.
