China import soybean from the United States,Brazil,Argentina and other countries has increased year by year,and these countries are mostly affected areas of dangerous fungal disease in soybean,in order to enhance the quarantine at the quality and efficiency and to prevent it spread to China,establish a rapid quarantine methods and practical processing technology is the key.This paper have researched Diaporthe/Phomopsis and Sudden death syndrome of soybean.Diaporthe/Phomopsis contain Diaporthe phaseolorum var.caulivora(DPC for short),Diaporthe phaseolorum var.rneridionalis(DPM for short),Phomopsis longicolla(PL for short ) and Diaporthe phaseolorum var.sojae(DPS for short),they always infect soybean together,and making it easy for misjudgment.The methods of morphology,pathogenicity and ITS(internal transcribed spacers) sequence analysis three aspects were used in this study,and have identified DPC,PL.At the same time, DPM,DPS standard strains morphological characteristics of PDA and internal transcribed spacer regions sequence have done.On this basis,DPC,DPM,PL Primers and TaqMan MGB probes were designed based on the nucleotide differences of DPC,DPM,DPS and PL within ITS,besides, have designed Diaporthe/Phomopsis common primers and probe.By repeating trials each have screen out one specific primers and probe,the technique of TaqMan MGB real-time fluorescent PCR was established to detect DPC,DPM,PL.The detected limitation were corresponding to 7fg/μl,6.5fg/μl,0.5pg/μl DNA.Besides,have sequencing intemal transcribed spacer regions and translation elongation factor 1-αgene regions to Fusarium cuneirostrum,F.brasiliense,F. virguliforrne,F.tucumaniae and F.phaseoli.Compared with GenBank has published sequence,and have fund Single Nucleotide different sites of this four interspecific(which did not find the differences sites between F.tucumaniae and F. phaseoli).Use SNP(Single Nucleotide Polymorphisms),Allelic Discrimination experiment,and have identified F.cuneirostrum,F.brasiliense,F.virguliforme,F. tucumaniae and F.phaseoli successfully.Achieved the purpose of detection. In the aspect of disposal technology,The toxicity of 8 fungicides to the DPC,DPM and PL were tested in laboratory.The result show that the toxicity of carbendazim 25%SC is the strongest to all the three kinds of pathogens,with EC50 values of 0.118mg/l,0.112mg/ml,and 0.152mg/l,respectively.Followed by difenoconazole 25%EC and hexaconazole 5%EC.So the three fungicides can be used for effective control of these diseases.
