The medical plant seed is the foundation of planting.The quality of seed good or not will affect the output and character of the product.In order to insure and improve the quality of seed,realize the quality standard of medical plant seed,we must carry out the quality testing of medical plant seed widely. The quality testing of medical plant seed has various contents,seed viability is the most important one among them.Nowadays,Chinese traditional medicine walks up to the outside world more and more.With the research development of the aging and inferior seed of crop and forest,the medical plant seed are paid attention too.Radix Astragali,Radix Codonopsis,Radix Bupleuri and Radix Polygale as the vital local officinal botany resource in Shanxi province,are all propagated with their seeds.Select the eminent seed is fairly pivotal when we develop the local Chinese traditional medicine and make the product territorially and sweepingly.On the other hand,examine the sample of the medical plant seed used advance science technology and international standard,then according to the results of which,we estimate the superior or inferior seeds of the medical plant and assess the value of them objectively, that are prodigious besteading for building the carrier of the excellent pharmacological character of the medical plant seeds.In order to illuminate the difference of the seeds of Radix Astragali, Radix Codonopsis,Radix Bupleuri and Radix Polygale especially in the aspects of viability in different years,we detected the leakage of electrolytes, antioxidant enzymes activities,malondialdehyde(MDA) and storage materials content and germination percentage by traditional germination technique.And also analyzed the relations between them by SPSS biostatistics software.Through the research we can not only discuss the realation between metabolize system and the viability in interior of that four species medical plant seeds,but also give a refer to assessing the quality of seed and setting up the critical database even the quality standard of medical plant seed.Primary investigation results as follow:(1) In the aspect of electrolytes leakage:the conductance rate of the seeds of Radix Astragali,Radix Codonopsis,Radix Bupleuri and Radix Polygale in different years,from total ascend to say,increased as the length of storage period increased.They all represent that the conductance rate of the new seeds<the conductance rate of the old seeds.Among of them, through the correlative analysis of the conductance rate and seed vigor,it was found that the conductance rate of the seeds of Radix Astragali was galactic significantly negative correlation with seed vigor.The coefficient is -0.871** (P<0.01).The conductance rate of the seeds of Radix Codonopsis was galactic significantly negative correlation with seed vigor.The coefficient is -0.993**(P<0.01).The conductance rate of the seeds of Radix Bupleuri was significantly negative correlation with seed vigor.The coefficient is -0.707* (P<0.05).The conductance rate of the seeds of Radix Polygale was negative correlation with seed vigor but not significantly.That is to say, mensurate the conductance rate can be considered a quick,useful checking method of the seed vigor of Radix Astragali,Radix Codonopsis,and Radix Bupleuri.(2) In the aspect of the activities of antioxidant enzymes:the activities of antioxidant enzymes of the seeds of Radix Astragali,Radix Codonopsis, Radix Bupleuri and Radix Polygale in different years,from total ascend to say, decreased as the length of storage period increased.They all represent that the activities of antioxidant enzymes of the new seeds>the activities of antioxidant enzymes of the old seeds.But the activities of catalase(CAT) of the seeds of Radix Codonopsis became abnormity.It represented that 2006year>2004year>2003year>2005year.Among of them,through the correlative analysis of the activities of antioxidant enzymes and seed vigor,it was found that only the activities of catalase(CAT) of the seeds of Radix Astragali was positive correlation with seed vigor.The coefficient is 0.765* (P<0.05).That is to say,mensurate the activities of catalase(CAT) can be considered a quick,useful checking method of the seed vigor of Radix Astragali.(3)In the aspect of the content of MDA:the content of MDA of the seeds of Radix Astragali,Radix Codonopsis,Radix Bupleuri and Radix Polygale in different years,from total ascend to say,increased as the length of storage period increased.They all represent that the content of MDA of the new seeds<the content of MDA of the old seeds.Through the correlative analysis of the content of MDA and seed vigor,it was found that the content of MDA of the seeds of Radix Astragali,Radix Codonopsis,Radix Bupleuri and Radix Polygale were all negative correlation with seed vigor but not significantly. That is to say,mensurate the content of MDA whether can be considered a quick,useful checking method of the seed vigor of Radix Astragali,Radix Codonopsis,Radix Bupleuri and Radix Polygale deserve discussing.(4)In the aspect of the content of storage materials:toward soluble protein,the content of soluble protein of the seeds of Radix Astragali,Radix Codonopsis,Radix Bupleuri and Radix Polygale in different years,from total ascend to say,decreased as the length of storage period increased.They all represent that the content of soluble protein of the new seeds>the content of soluble protein of the old seeds.Toward soluble sugar,the seeds of Radix Astragali,Radix Codonopsis and Radix Polygale all represent that the content of soluble sugar of the new seeds<the content of soluble sugar of the old seeds.But the content of soluble sugar of the seed of Radix Bupleuri became abnormity.It represented that 2004year>2003year>2005year>2006year. Among of them,through the correlative analysis of the content of storage materials and seed vigor,it was found that the content of soluble sugar of the seeds of Radix Astragali was galactic significantly negative correlation with seed vigor.The coefficient is -0.894**(P<0.01).The content of soluble sugar,protein of the seeds of Radix Codonopsis was galactic significantly negative and positive correlation with its germination rate.The coefficient is -0.814**,0.969** respectively(P<0.01).Besides,the content of soluble sugar,protein of the seeds of Radix Bupleuri and Radix Polygale was negative or positive correlation with their germination rate,but not significantly.That is to say,mensurate the content of storage materials can be considered a quick,useful checking method of the seed vigor of Radix Astragali,Radix Codonopsis.(5)Through dyeing the seeds of Radix Astragali of three different years to test their vitality by TTC and germination percentage by traditional germination technique,we get the results that the highest average vitality level is the seeds in 2006year,about 73.33%.But the highest average germination percentage is the seeds in 2004year,about 26%.The two conventional and classical checking method of the seed vigor can not become solidified or united.So through dyeing the seeds to test its vitality by TTC whether can be considered a quick,useful checking method of the seed vigor of Radix Astragali deserve discussing.Moreover,in view of limitation of TTC,the seeds of Radix Codonopsis,Radix Bupleuri and Radix Polygale were not taken into this experiment.(6)In the aspect of the seed germination:the average germination percentage of the seeds of Radix Codonopsis,Radix Bupleuri and Radix Polygale in different years,from total ascend to say,decreased as the length of storage period increased.They all represent that the average germination percentage of the new seeds>the average germination percentage of the old seeds.But average germination percentage of the seeds of Radix Astragali became abnormity.It represented that 2004year>2006year>2005year. Finally,we regard mensurating germination as the most efficient and credible checking method of the seed vigor of Radix Codonopsis,Radix Bupleuri and Radix Polygale.But toward the seeds of Radix Astragali whether can be considered an efficient and credible checking method also deserve discussing.
