The production of CLA in sterile and natural small intestinal fluid was studied by fermentation with Lactobacillus plantarum ANCLA01.Using extrasomatic method,this experiment studied the influences of lypase,sunflower seed oil,inoculum size,pH and time in fermentation,which regulated the conditions for producing CLA.The results are as follows:Experiment One:effects of different raise condition on the production of CLA by sterile small intestinal fluid with Lactobacillus plantarum ANCLA01After sterilization at high temperature,mixed the fresh small intestinal fluid with prepared nutrient solution by 1:1,put the mixture in a 50 mL centr fuge tube with screwed cap and added 100U/mL lypase,sunflower seed oil,Lactobacillus plantarum ANCLA01.Then,twisted the cap after excluding the air by pure CO2,and used plastic bag to wrap centrifuge tube,kept fermenting at 37℃.The results showed that adding lypase could obviously produce more CLA;Optimal fermentation conditions were indicated: sunflower seed oil volume was 3.5mg/mL,3%inoculum size,pH was 6.5,fermentation time was 28h,the yield of conjugated linoleic acid was 55.78 ug/mL.Experiment Two:effects of different raise condition on the production of CLA by natural small intestinal fluid Lactobacillus plantarum ANCLA01The fresh small intestinal fluid mixed with nutrient solution by 1:1,the method was the same with that of Experiment One.The results showed that adding lypase could obviously produce more CLA;Optimal fermentation conditions was indicated:sunflower oil volume was 3.5mg/mL,4%inoculum size,pH was 6.5,fermentation time was 32h,the yield of conjugated linoleic acid was 51.47ug/mL.
