Taxus culture cell is one of the solutions for exhaustion of Paclitaxel resource problem. The general material of producing Paclitaxel is plant tissues, and there is adaptive process for it. As the fast developing of separating technology and the changing of materials, the existing process has showed several facts inefficient.Based on the modern chromatographic theory,and the existing manufacturing method, the present research focus on developing an adaptive process for producing Paclitaxel from the culture cell material, and with the advantages of low cost, high yield, etc. The details are as follows.1. Used different molecular scale membrane to separate the target substances. The process could efficiently compress the volume of cell material, prevent Taxol loss out of cell, and discard the high polar foreign matters. This is key to save the organic solvent for later processes.2. Selected a resin type and establishing a new separating process by adaptive eluting system after the membrane separating process. This new process has raised the Taxol concentration up to 0.8%, and has the advantages of low-poisoned solvent system, mass sample loading, good recovery of media. The process has the potential replacing the current L-L extracting process.3. Established a process with positive and reverse phase chromatography, it can effectively improve the purity up to 85%, with the 70% recovery. The whole process is oriented to large-scaled manufacture, and the experiment has made a foundation for further research.
