To meet the improvement and the development of intensive animal husbandry, bulls were on keeping to production of Frozen semen,which play an important role in the animal husbandry.At present,the urgent necessary to sort out our active bulls resources and genetic resources,collect and collate their Family performance and pedigree information of the relationships,and to build the database of bulls to map out the clearly reflected map of the relationships of the bulls pedigree are the premise and basis of the introduction of scientific,scientific matching and cattle breeding. Therefore,based on the evaluation of the genetic diversity,studying and developing the bulls individual identification techniques and methods have a great significance to the healthy and sustainable animal husbandry.In this paper,according to the United Nations Food and Agriculture Organization (FAO) and the International Fund for Animal Genetics(ISAG),the recommended 14 pairs of high polymorphism microsatellites primers were selected,and then based on the ABI3100-automatic sequence analysis,collected the 330 bulls individuals from the north of China,using PCR technology to amplify TGLA227,BM2113,TGLA53, ETH10,SPS115,TGLA126,TGLA122,INRA23,ETH3,ETH225,BM1824 11 pairs of chromosomes STR loci and three pairs of Y-chromosome STR loci UMN0108, MCM158,MAF45.then the amplified PCR products were tested throught the polyacrylic acid gel amine electrophoresis(PAGE) and detected by silver staining after polymorphism distribution,and then the capillary electrophoresis instruments. And calculation of the 14 loci gene frequency(Pi),Individual identification Potential (DP),heterozygosity(H),the polymorphic information content(PIC),Cumulative Individual identification Potential(TDP) and non-paternity exclusion(PE).The results are as follows:1.14 STR loci are highly genetic polymorphism,and among the dairy cattle,a total of 133 alleles have been detected,with an average 9.5 for each seat.In the beef cattle,a total of 137 alleles have been detected,with an average 9.8 for each seat. Among the 14 polymorphic microsatellites loci the PIC between 0.888-0.482,the average dairy cattle is 0.804,while the beef cattle 0.735.and we found that the average heterozygosity(H) relatively higher than that of beef cattle in China.Finally, the genetic diversity detections showed that north and northeastem China bulls have the power of potential choices and relatively abundant genetic diversity.2.the gene frequencies differ from the different STR loci in both cattle,and the special genes were detected in total 41.While,those 14 STR loci genotypes distributions are not consistent with Hardy - Weinberg equilibrium.It is obviously consistent with the fact that the bulls experienced the high artificial intensive choices.3.the results of the feasibility of the application to individual identification and paternity testing showed that those 14 STR loci value of the individual identification Potential(DP) raged from 0.696 to 0.968,which the MCM158 locus has the highest DP value:0.968,while the loci SPS115 has the least DP value 0.696.And the Cumulative Individual identification Potential(TDP) both reached 99%,and the Cumulative Non-paternity Exclusion(PE) also reached 99.99%,both meet the need of medical jurisprudence.Which also proved that those 14 microsatellites loci can be used for dairy,beef cattle individual identification and the pro-identification.
