Research On The Histological Structure Of Rana Nigromaculata's Organs And The Histochemistry Of The Frog's Mast Cells

Objiective:To research the histological structure of Rana nigromaculata's organs and the histochemistry characteristic of the frog's mast cells(MC).Methord:Three kinds of stationary liquid,10%formaldehyde solution,Bouin fluid and Carnoy fluid,three staining ways,hematoxylin-eosin(HE),modified toluidine staining(MTB) and alcian blue-safrianin O(AB/SO) were used to study the histological structure of Rana nigromaculata's organs and the histochemistry of the frog's mast cells.Results: 1.The histological structure of Rana nigromaculata's lung,spleen,liver,stomach, intestine pancreas,kidney,testicle and ovary was described.Spleen was composed with red pulb and white pulb,but periarterial lymphatic sheath was not seen in the white pulb.The structure of lung was simple,the epithelia of alveolus included cuboidal epithelium and squamous epithelium.Some lymphocytes or lymphatic nodule could be seen in some organs of Rana nigromaculata,they played a role in immunity.2.MC of Rana nigromaculata could be showed by staining with using MTB or AB/SO when fixing with each fixative.Bouin fluid was fine stationary liquid to display the Rana nigromaculata's MC.3.The distribution and shape of MC in different organs of the frog was diverse.The cells mainly distributed in the skin, tongue,stomach,intestines and spleen of each frog,especially a lot of mast cells coulde be seen in the stannius corpuscles.The number of MC in the heart,liver, kidney,testicle and ovary of frogs was seldom.4.Fixed with Bouin fluid and stained with MTB,three kinds of MC could be seen in the tongues of frogs.The grain in some MC was pink,the grain in some MC was blue,and there was pink grain and blue grain in some MC.But the grain of MC distributed in the other organs was blue. 5.MC around the cyst in the wall of small intestine attacked by parasite increased, and the cells appeared some heterogeneity.Conclusion:Similarities and differences existed between the histological structure of Rana nigromaculata's organs and that of other batrachian.MC could be seen in each organ of Rana nigromaculata,Bouin fluid was fine fixative solution to display the MC of Rana nigromaculata when using MTB or AB/SO.The distribution and histochemical characteristic of Rana nigromaculata's MC were different from that of other batrachian.MC played a part in anti-infection of parasite.
