The fungal biocontrol agent Paecilomyces lilacinus,was evaluated for its potential to control the plant-parasitic nematodes on series of plant species.Study on the effect of the plant species on the persistence of Paecilomyces lilacinus strain 221 in soil and the rhizosphere were investigated.The original strain of the Paecilomyces lilacinus 36-1 with the drug resistance of carbendazim at the concentration of 2.0μg/ml.The drug resistance fungal strain 221(PL221) was mutated by ultraviolet and the the resistance level to carbendazim was 30.0μg/ml.As the target strain PL221 is better than original strain 36-1 on the biological characteristics such as sporulation ability,spore germination rate and ability of kill nematode.It's important to select efficient isolation media on the target strain recovery.The OHIO-agar out of the five medias(OHIO-agar,Gause's,Czapek's,PSA,Martin's)was the best media for isolation the Paecilomyces lilacinus from the soil and plant rhizospheres.Paecilomyces lilacinus strain 221 in soil can survival more than 60 days.The strain were applied into soil in the way of solid-state fermented culture and spore suspension, the application methods of solid-state fermented culture have higher biomass of the survival.Studied the colonization of PL221 in the pot trail and field by different crop,different genotype of crop,different soil type and strain application method.It provides preliminary knowledge of the difference of colonization in plant foundation soil, rhizosphere and endophytic root.In addition,the biomass of the detection has a certain negative correlation between average soil temperature and precipitation.PL221 can be colonized in plant foundation soil,rhizosphere and endophytic root in different 5 plants in pot trail.The biomass of plant foundation soil is above 10~6 of magnitudes,there was the high biomass on cron,tomato,cowpea,Steady of the colonization biomass in plant rhizosphere of the tomato and cucumber with the biomass above 10~5 of magnitudes,and the biomass of endophytic root between 10~2 and 10~4.9 crops in field were detected.PL221 can be detected in the plant rhizosphere, endophytic root and the plant foundation soil which the biomass between 10~4 to 10~6 of magnitudes,and the rhizosphere above 10~4 of magnitudes.The colonization ability of the PL221 in plant foundation soil of corn in field was worse than the other,tomato and cucumber were good.PL221 can be colonized in plant foundation soil,rhizosphere and endophytic root of the 5 different genotypes of tomato and cron.The biomass of plant foundation soil is high than the rhizosphere,the third is the endophytic root.The average biomass of plant foundation soil is about 10~6 of magnitudes,the rhizosphere above 10~4 of magnitudes, discontinuities of the determination in the plant endophytic root.The colonization ability of the TEDA genotypes of tomato is better than the other.Steady of the colonization biomass in plant foundation soil and rhizosphere of the cron inbred line.PL221 can be colonized in plant foundation soil,rhizosphere and endophytic root in tomato(TEDA) and cotton(ERMIAN 23) in 3 different soil types,there are alluvial soil(HN),yellow soil(HB),yellow brown soil(GX).The biomass of plant foundation soil is high than the rhizosphere,discontinuities of the determination in the plant endophytic root.The biomass of plant foundation soil is above 10~6 of magnitudes,the rhizosphere above 10~4 of magnitudes.The 4 strain application methods(seed soaking,root dipping,spore suspension and solid-state fermented culture treat the soil) were applied into tomato soil in pot.PL221 can be colonized in plant foundation soil,rhizosphere and endophytic root in tomato (TEDA).The good colonization effect of plant foundation soil and rhizosphere by the strain application methods of spore suspension and solid-state fermented culture,the biomass of colonization is above 10~6 of magnitudes.The strain application methods of root dipping was the best one for the strain colonized in plant endophytic root.Seed soaking was not good in the 4 strain application methods.Tissue isolation of the plant root and stem can be detected the target strain in the colony edge.It shows that PL221 can colonization in plant root.It also can be detected in the leaf of plant,preliminary proof of the PL221 can colonization in plant stem and leaf.
