Object: In order to gain the high purities and concentration of proteinfrom ginseng and deer antler, a foundation for ginseng and deer antler'sstudy of structure and function, a simple and efficient purification methodwas established.Method: The aggregate proteins were purified by means of salt solutionextraction, ammonium sulphate precipitation, ultrafiltration, affinitychromatography, iron exchange chromatography and hydrophobic interactionchromatography; The purity of the purified protein were identified bySDS-polyacrylamide gels electrophoresis and gel filtrationchromatography.Result:1. Five ginseng proteins and four deer antler proteins were purified from ginseng and deer antler, respectively, by means of established corresponding purification protocol. All the purified proteins showed single band/or peak after SDS-PAGE and HPLC.2. The receival percentage of purified protein of deer antler was above 75%, while that of GP1 and GP2 in ginseng was above 80%.3. The purified proteins of ginseng, generated as GP1, GP2, GP3 and GP5 were made of one single subunit, with 25kD, 65kD, 15kD and 8kD in molecular weights, while that of the GP4 fraction was hetero-dimmer glycoprotein with relative molecular weight, 29kD and 27kD, respectively.4. The purified deer antler proteins were generated as DAP-1, DAP-2, DAP-3 and DAP-4. DAP-1 was heterogeneous tetramer, consist two kinds of subunits (50kD and 27kD) owning the molecular weight of 140kD; while DAP-2 was isogenous dimmer owning the molecular weight of 120kD, 64kD for each subuint; DAP-3 was also isogenous dimmer owning the molecular weight of 28kD, whose subunit's molecular weight is 13kD while DAP-4 was a 6kD single subunit protein.5. The nature of purified nine kinds of proteins, GP1 and GP3, GP4, is ribonuclease-like protein and chitin protein, respectively; GP2 and GP5 of ginseng protein and four kinds of deer antler protein, DAP-1 to DAP-4, were first reported, their properties need to investigate later.Conclusion: Aiming at two important Chinese traditional medicine—ginseng and deer antler and studying the high concentration of protein in them, this research adopted advanced biological technology to isolate and purify nine kinds of proteins. We conquered the phenomenon of low standard of technology and the immoderate protocol and so on, then we constituted a purified protocol which was applied to many sorts of proteins. The result indicated that this protocol was fast, simple and efficient.
