Leymus is a potentially useful source of a number of genes for wheat improvement, including powdery mildew resistances, rust resistance, drought tolerance, an increased number of florets per spike. Octoploid Tritileymus is an incomplete amphiploid with Triticum and Leymus. It's an excellent bridge materal which is used to transfer good Leymus genetic Material. In this study, we using of molecular cytogenetics and molecular biological methods for the identification of hybridization between Triticum and Leymus. Cytological analysis was contact to xiaobin950059, which is Octoploid Tritileymus, and hybrid between xiaobin950059 and other material. Feasibility analysis of transferring the Leymus gene into Triticum by pollen-tube pathway method. The results are as following:1.The morphological trait and cytogenetic characters of two leymus/Am6-4 F3 (N06212-1 and N06212-2) were identified, they all had the Chromosome constitution of N06212-1 and N06212-2 were confirmed by in situ hybridization. N06212-1 had 43 wheat chromosomes and 1 leymus chromosomes, and N06212-2 had 42 wheat chromosomes and 3 leymus chromosomes.2. Cytological characteristic and in situ hybridization were studied on Meiosis and Pollen development of xiaobin950059, it had 22 pair of wheat chromosomes and 6 pairs of leymus chromosomes. And in the process of microsporogenesis and male gametophyte development of Tritileymus,some abnormal phenomenon were observed.3. The process of the meiosis of pollen mother cells and pollen development of hybridization between Yingcumai, Am6-4, Jinsong5hao, TE256, Xiaoyan7631, Yannong15 and Xiaobin950059. The chromosome configurations of these hybridization were different with theoretical value. And we were discussed the genetic relationship between various chromosome set.4. Cytological characteristic and in situ hybridization were studied on octoploid Tritileymus/Am6-4 F3. We identified two additional lines (N06129-4 and N06130-10) which had additional two leymus chromosomes and one leymus chromosomes. They can be used for the development of substitution line and translocations.5. Introduced leymus DNA into common wheat cultivar China spring, and cytological characteristic and in situ hybridization with these China spring. The results showed that three pairs of SSR primers(Xgwm159-5B,Barc163-4BL,Barc178-6BL) who showed polymorphism in seven plants. Seven plants had appeared the chromosome aberan ces which including single chromosome,chromosome lagging,chromosome bridge,excessive bally dividing etc. Only two plants appeared in situ hybridization signals. It showed that these China spring had changed in the genetic composition.
