Nuogu pig and luobo pig are both excellent species with wide representativeness in Guizhou province,featured fresh meat quality,good mouthfeel,crude feeding and great disease resistance.These pig species come into being by long time natural and manual selection in nature entironment.But because of the slowly growth,low lean meat percentage and the lower economic interest comparing to the species introduced from other countries.Now the Nuogu pig and Luobo pig decrease sharply to the border of extinction.The genes regulated to the excellent meat quality traits of the two species are not clearly explained.Choosing the two species,Nuogu pig and Luobo pig,as the study subject,this research has investigated the meat quality traits, carcrass traits and the mechanism of molecular heredity,to make a scientific use of the Guizhou pig species in pig production,and select some available molecular markers for the meat quality improvement.The result as follows:The study's subject:Nuogu pig,from Nuogu production study institute at NaYong country in BiJie region of Guizhou province;Luobo pig from JiangKou hoggery in TongRen region;and the commodity pig from the slaughterhouse of HuaXi in GuiYang.Using the PCR-RFLP and PCR-SSCP to study the candidate genes UCP3 and H-FABP,the research analyzes the effect of the two genes' mutation on the meat quality traits of GuiZhou local pig species,which provides a theoretical basis for ascertaining the major gene affecting the intramuscular fat sediment and its closely related hereditary marker.The result suggests:Making the PCR amplification outcomes of the 6th exon 167bp of UCP3 gene and the 2nd intron 816bp of the H-FABP analyzed by PCR-SSCP and PCR-RFLP,a mononucleotide polymorphism mutation situs and a HinfI-RFLP polymorphism situs are discovered respectively,which are basically accordant with literature has reported.Out of the GuiZhou regional pig species,the 6th exon of UCP3 gene and the 2nd intron of the H-FABP have the polymorphism.The 6th exon of UCP3 gene has three genotypes:AA,AB and BB,their respective frequency are in Nuogu pig 0.4333, 0.5000 and 0.0667,in Luobo pig 0.3400,0.6200 and 0.0400,in hybrid breed 0.0833, 0.2834 and 0.6333;The 2nd intron region of the H-FABP gene has three genotypes too: HH,Hh and hh,and their respective frequency are in Nuogu pig 0.4333,0.1000 and 0.4667,in Luobo pig 0.2000,0.4400 and 0.3600,in hybrid breed 0.2667,0.2667 and 0.4666.In the two polymorphism situs,the difference of the hereditary traits among the there pig groups is also obvious.The UCP3 genes of the three pig groups reveal a moderate polymorphism all(0.25<PIC<0.5),and the H-FABP gene also reveal a moderate polymorphism in three pig groups(0.25<PIC<0.5).The effect of the the H-FABP gene on the area of longissimus muscle,meat color, drip loss,pH 1 value,pH 24 value,cooking loss,marbling score,intramuscular fat content and tenderness is not significant(P>0.05);the effect of the UCP3 gene on the marbling score,intramuscular fat content and tenderness is significantly different(P<0.05) while on other traits are not.
