Tibial dyschondroplasia is a cartilage metabolic disease which is wide spread in poultry industry,with high incidences in young broilers,laying chickens and turkeys.The TD chickens are clinically characterized by mental depression,reduced appetite, groveling on the ground usually,dyskinesia,and have difficulty to stand,sometimes walking with wings supported on the floor when seriously.The TD epiphyseal growth plate becomes thick,sometimes forms opaque uncalcified and unvascular wedge-shaped cartilage extending from epiphyseal to metaphyseal.Tibial dyschondroplasia can not only reduce chickens disease resistance,performace trait and chicken quality;but also induce chest-cystis and carious osteitis,and cause great loss to poulty industry.Based on success of tibial dyschondroplasia introduced by thiram in vitrol,healthy broiler growth plate chondrocytes were cultured in vitrol to study the effect of thiram on alkaline phosphatase,acid phosphatase and nitric oxide synthase and expression of Matrilin-3 gene related to bone mineralization.The thiram concentrations in different groups are(0.1,1,3,5μmol).After 72h,results are assayed.The result showed that compared with control group,the activities of alkaline phosphatase and nitric oxide synthase increased as the concentrations of thiram increased. The semi-quantitative RT-PCR result showed the expression of Matrilin-3 had a down regulation as the concentrations increased.Thiram affected enzymes activities related to development of tibial chondrocytes and reduced the expression of matrilin-3 gene,which is the etiology of tibial dyschondroplasia cauded by thiram in broilers.
