Polyploid species of plants plays an important role in plants evolution.It is estimated that at least 50%or even more than 70%of the angiosperms was diploidizated from it's ancient.Wheat(Triticum Aestivum)which is hexaploid with 3 different genomes,has strong tolerance for related species' genomes ingress.So it is easy to Synthetic wheat and wheat related species amphiploids.Those amphiploids not only can serve as breeding resources,production of a variety of applications,but also be used to study the interaction genome,genome evolution,and epigenetic variety.Wheat seed storage protein is an important component of wheat seed and is closed related to wheat quality.The predecessors have studied on the gene sites of wheat seed storage protein and its expression deeply.Rye is a very important resources to resistance to powdery mildew,amphiploid between wheat and rye was synthesized to utilize.Two types of amphiploids:between Chinese spring(CS)-Rye AR106BONE(AR) was abbreviated as CA,MianYang11(MY11)-Rye AR was abbreviated as MA which were identifyied by GISH,SDS-PAGE and A-PAGE and evaluated its resistance to powdery mildew.Results are as follows:1.GISH identified two sets of a total of 20 materials and the mid-chromosome number is 2n=56 in cell meiosis.14 of them was determined as Secale's chromosome.SDS-PAGE and A-PAGE showed most of parents' bands which means that:we got the amphiploids successfully.2.Several generations had varietied from its parents for some bands of parents lack,as well as other new bands which not showed in parents.The slower band of Secale AR had lacked both in CA and MA and subunit 2of CS did not exsit,but near to subunit 7 there were more than 2 new bands in CA.All subunits of MY11 presented in MA.That indicated there were differences intergenomic interaction in different doner wheat in the aspect of HMW-GS. 3.HMW-GS and gliadin have big differences in inhertting stability in generations.The 20 materials have the same HMW-GS electrophoresis pattern,while have big differenced between gliadin banding patterns,That means HMW-GS may be more stable than gliadin in heredity.The polymorphism of MA is richer than CA which may showed CS have stronger containing ability to alien chromosomes and may be easier to be hybridized for getting amphiploid4.High generation rye AR disappeared sacalin protein,while its generations contain or partly contain secalin protein bangding.That showed there are strong changed on Sec-1 of 1R chromosome.We should pay more attention on the principle of expressing and controlling abilities of Sec-15.When inoculated with powdery mildew isolates,two types amphiploids both expressed strong resistance derived from its rye parent.
