Zea mays is of mays of Zea of Gramineae. It is C4 plants, and it has advantages including high-yield, industrial chain being long, yield potential being large, so maize holds an important role in agricultural production. Currently, maize is one of three major food crops of the world, having been second largest food crops in China, and it is important raw materials to food ,medicine, and other industry. Maize occupys total seed 8% to 12%, and its protein content is about 20%. But maize embryo usually as waste disposal is discarded.In this study, experiment material is series of NEI DAN maize seed. The methods include half-dry extract embryo, drying, coarse grinding, skim, second smash, hydrolysis using delicate neutral enzyme and complex protein enzyme, Poly Acrylamide gel electrophoresis, ultrafiltration, debittering, concentrate, drying and so on.The results of this study show that: the protein content of the unskimed embryo samples of maize seed selected series of NEI DAN is 22.71%, belongs to higher protein content, and this variety is suitable for raw materials of comprehensive utilization and in-depth processing of maize embryo. The rate of using salt-soluble extract protein is low, loss is large, unsuitable for product polypeptide of maize embryo protein. And delicate neutral enzyme and complex protein enzyme hydrolysis the production, getting polypeptide elements which molecular weight focused on 200~2000Da after ultrafiltration, can meet experimental requirements. The optimum conditions of hydrolysis are as follows: the concentration of substrate is 3%, the concentration of delicate neutral enzyme is 1‰, the concentration of complex enzyme is 2.5‰, respectively, and hydrolysis 3 hours in its optimum pH and temperature conditions.
