For the past few years, with the extensive development of the cows industry, the cow dermatomycosis happened year by year. It not only has negative impact to cows industry but also affect people's production.The study objects were cows with dermatomycosis, 28 patients were collected and isolated cultured. 17 cows had separated trichophyton verrucosum, and provided a consult for cows'nosomycosis study. Drug sensitivity tests of the bacteria were taken (Main reference NCCLS M38-A programme), ketoconazole and itraconazole were chosen to take invitro inhibition test. The results proved that itraconazole was more sensitive to the bacteria, and provided a reference for itraconazole clinical application. The research of itraconazole dosage form was taken. On the basis of previous research, itraconazole hydroxypropyl-β- cyclodextrin was prepared successfully. The corresponding experiments had been taken, and the results proved that this preparation could be used in veterinary surgeon clinical.Proper dosage regimen was provided for itraconazole clinical application and the research of itraconazole pharmacokinetic in goats has done. Itraconazole injection was injected into goats intramuscular by 4mg/kg, and collection blood by vein after 0.5,1,2,3,4,5,6,7,8,9,10,11,12,24,48 hours using heparin anticoagulation. Methanol precipitation protein, high-speed centrifugal separation method was used to purification sample. High performance liquid chromatogram-violet detect method was used for concentration determination, and the results were analysis by the MCPKP pharmacokinetic software. The limit of detection for itraconazole was 20ng/mL, and a good linearity was obtained in the range of 20-1000ng/mL. Recovery rate and precision were permitted within the scope of the method, and the method was simple, low cost, non-interference, suitable for clinical application. The pharmacokinetic parameters of itraconazole injection were as follow: the apparent volume of distribution 0.05843L/kg; a excretion rate constant 0.02020±0.01092/h; drug half-life 34.31289h; the area under the plasma concentration-time curve 0.33898×104 mg/(L·h); plasma clearance rate 0.0018 L/(kg·h); plasma concentration time 357.59h; maximum drug concentration 226.67ng/mL; the time to get the highest concentration 3h. The itraconazole pharmacokinetic process in goats was one-compartment model. The theoretical support for clinical effective and rational use of itraconazole rejection was provided. The invitro inhibition tests of trichophyton verrucosum were studied and have achieved good results. Meanwhile, the pharmacokinetic characteristics of itraconazole intramuscular rejection in goats were studied for the first time, and corresponding results were made. Theoretical foundation was established for itraconazole rejection clinical application in cows'nosomycosis and contributions have made to purify cows'nosomycosis, ensure milk source security and public health security.
