The locoweed is one of kind poisonous plants that endangers the husbandry seriously in the world, and causes enormous economic losses. Many measures such as employing weedicide or herbicide and special machines, were adopted to remove locoweeds from sources of exposure, and these ways were currently major approaches to protect animals from locoweed attack in China, but still now, there is no effective way for curing locodisease. The main aim of this study is to design optimal immune program, gain the serum against swainsonine with high titer, cure the locodisease of goats, evaluate and analysis indexes of serum against swainsonine treated on goats poisoned by O. kansuensis Bunge, study the effects of serum against swainsonine treated on goats poisoned by O. kansuensis Bunge. The results are as follow:1. After poisoned, several enzymes were changed caused by toxin on the day 7, light clinical symptom was observed on the day 14, and typical poisoned symptom presented on the day 21. Model goats poisoned by O. kansuensis Bunge had been constructed successfully.2. On the day 35, antibody titer presented, and the level of antibody titer had reached the highest on the day 42, someone was 213, however, there was no significant difference between the day 42 and the day 49. After the day 49, blood was collected from every rabbit by arteria carotis, and 135 mL serum against swainsonine which titer was over 210 was gained totally.3. The total of white blood cell in all goats was not presented significant difference, however, there was significant difference between poisoned and cured group (group B) and poisoned and control group (group C) in red blood cell number. Lymphocyte number and neutrophil number were as red blood cell number. These suggested that serum against swainsonine may inhibit the anaemia caused by swainsonine and protect the hematological system from injuring.4. From the change of serum enzymes, on the day 42, LDH levels of group B was extremely significant lower than that in Group C, but compare with the control group (Group A), was still higher. On the day 28, there were an extremely significant difference among three groups, on the day 35, the serum AKP levels of Group B was extremely lower than Group C, but still significant higher than that in group A. on the day 42 to the end of experiment, the serum AKP levels in all group existed a significant difference. On the day 28, the activity of Serum AMA in Group B was significant higher than that in Group C, on the day 35 there were an extremely significant difference among the three groups. From the day 42 to the end of this experiment, serum AMA level in Group B was significant higher than that in Group C. On the day 35, there was significant difference among three group in the activity of serum BUN, From the day 42 to the end, the activity of Serum BUN in group B and Group C were extremely higher than that in group A(P<0.01), but there were no significant differences between group B and group C. These suggest that serum against SW may neutralize the swainsonine and delayed or rectified the change caused by locoweed.5. Serum against swainsonine could slow down the speed of E-rosette rate of decay, and protect the immune system from injuring.6. On the day 35, the change about concentration of swainsonine between group C and group B were significant difference, this suggest serum against swainsonine could neutralize swainsonine partially.7. From the detection of Histopathology, the results suggested vacuolization and Granular degeneration in Group B was light than that in Group C. |
