| In this research,a wheat recombinant inbred line population including 111 F2:6 families,which derived from the across between WSSMV resistant and susceptible varieties,Xifeng and Zhen 9523,was generated by single seed descent(SSD).A total number of 709 primer pairs mapped on 21 wheat chromosomes were employed randomly to reveal the polymorphism between two parents.295 primer pairs of which were polymorphic,and the polymorphic rate was 41.6%.Further analysis of 148 polymorphic primer pairs in the selected 111 families indicated that the mean of polymorphic index(PI) was 0.5,ranging from 0.40 to 0.50.Segregation ratio of 126(85.1%) markers conformed to the 1:1(P>0.05),while 22(14.9%) markers showed segregation distortion in the tested families.This indicated that genetic contributions from the two parents are closely equal. These results suggested that the RIL population has reasonable genetic composition and should be ideal for wheat genomic researches such as genetic mapping,molecular tagging and molecular breeding.Eight-seven markers were further used for linkage map analysis using Mapmaker 3.0 software.Twenty-four linkage groups,which covered 1204.3 cM of wheat genome with the average genetic distance standing about 13.8cM,were constructed.According to the reported chromosome location of the analyzed molecular markers,these 24 linkage groups belonged to chromosomes 1D,2B,2D,3A,3B,3D,4A,4B,4D,5A,5B,6A(6B), 7A,7B and 7D,respectively.Based on the genetic map constructed,QTL analysis was performed using joint composite interval mapping using the CARTOGRAPHER2.5 software.Seven QTLs related to WSSMV resistance of Xifeng were detected on chromosomes 3B,2D and 4A.The contribution rate of QTLs located in chromosomes 4A were the highest,which were 12.0%. |
