The factors which affected the efficiencies of microbial purification were studied by the UV uncircle depuration system in this research,including environment factors such as shellfish to water ratios,water exchange rates,temperatures,salinities and an inner one as initial shellfish quality.The results showed that the purification efficiencies were strengthened while the shellfish to water ratios and water exchange rates increased.Tegillarcula granosa could be effectively purified during 13~30℃and the most effective temperature was 28~30℃, reflecting that T.granosa could endure the high and wide temperatures.However,the suitable temperature range for Crassostrea.Plicatulam's purification was 15~30℃,and the most suitable temperature was 25℃.The suitable salinity was 15~30‰not 10‰and the best salinity was 25‰,reflecting these two shellfish's specificities of enduring the low and wide salinities.The results showed that the shelflives of modifed atomosphere packagings of the live T.granosa storing under the temperatures of 0℃,4℃and 10℃were 17,13,7days while the air packaging was 7 days storing at 4℃and 6 days shorter than the MAP ones under the same condition.The shelflife could be much more longer while MAP was combined with low temperature.During the shelflife of MAP T.granosa,the changes of main nutritional compositiones were not significant,while the flavor was maintained.Modified atmosphere packagings of purified T.granosa were stored under the temperature of 0℃,4℃and 10℃.The Kinetics Models of total bacterial counts and TVB-N were developed based on these two changes by the temperature and time during the shelflives,so as to predicate and control the quality of T.granosa during storage.The changes of TVC and TVB-N value with time and temperature showed precision of the first order chemical reaction model and Arrhenius equation.A high performance liquid chromatography method for determination of ATP,ADP,AMP in bivalves was studied.This method showed high speed,accuracy and easy performace.The calibration curve showed a good linearity of theln(r2>0.9995) and the recoveries of ATP,ADP,AMP were ranged from 85.73 to 106.16%.Their precisions were lower than 10%while the limits of detection were 0.023,0.008,0.058μg·mL-1.The verified results demonstrated that this method was precise,accurate and can be used for determination of ATP in different bivalves' tissues.The establishment of this method can provide keys to judge whether the shellfish is live or not when it is dormancy.
