The garlic is one of important vegetable crops Xinjiang. The virus disease with stripe mosaic,stunt,distorting leaf , necrosis effect on the production and quality of garlic in North XinJiang. In order to make clear the sorts of virus in garlic virus disease , in 2007,garlic virus diseases main regions were investigated,in Urumchi,Shihezi,Jimusaer,Yilizhaosu and 76 state farm and so on. By electron microscope observation ,the virus particles that is 780×11nm were observed.Some special primers were designed based on generally reported virus gene sequences in garlic ,only a fragment of about 1402bp was amplified by RT-PCR using the total RNA of the garlic infected by OYDV special primer. The products of PCR were cloned into PMD18-T and transformed into TOP10. Through clony PCR detection, the fragments obtained are about 1402bp. The five combined clones were sequenced, and analyzed by DANMAN soft , the result showed that the fragment is 1402bp, encode 467 amino acids . The five clones form two groups, the obvious difference were seen between the two groups. In one group , including XJJM-3 and XJJM-6, had the highest nucleic acid sequence identity 99% and amino acid sequence identity 98% with AJ409311 from Shandong province Jinxiang of China. The other group consisted of XJJM-2, XJJM-4 and XJJM-5, the homology could reach 99%of nucleic acid and 99% of amino acid, compared with AB000481 from Japan, the result showed that the homology could reach 98%of nucleic acid and 98%of amino acid.
