During the process of tooth development,Wnt is one of the four kinds of growth factors transmiting inducing signals.However,the exactly role of Wnts on the tooth development is not clear.This study attempted to research how sfrp2 regulates Wnt signal system so as to predict the exactly role of Wnts on the process of tooth development.By means of molecular biology related experiment technique,we constructed respcectively the lentiviral transport vector,pNL-sfrp2-IRES2-EGFP, composes the full-length sfrp2 cDNA and pNL-U6-shsfrp2-EGFP composes the U6-shsfrp2,RNAi expression frame.Combinding them respectively with pHelper and pVSVG co-transfected 293T cells to produce HIV-1 virus.We infected mouse molar mesenchyma by these virus respectively to up-regulate or down-regulate the expression level of sfrp2.After detecting the expression level of sfrp2 by real-time PCR we check up the tissue infected by lentivirus.The result show the tooth development process will be postponed while up-regulating the expression of sfrp2.On the contrary,the process will be expedited while down-regulating the expression of sfrp2.These result demonstrated the Wnt signaling plays an important role in early tooth developmemt...
