Investigation of root-knot nematode disease was carryed out in 13 cities/counties include Jinling, Tanluo, Pubei, Lingshan, Tiandong, Tianyang, Yulin in Guangxi. The disease occurred in all areas investigated. Detection rate of second-stage juvenile(J2) was beteen 50% to 100%. Population density was between 14.8 J2 to 321.8 J2 in 100 ml soil. Root-knot nematode disease was found at all banana varieties investigated. Detection rate of J2 was between 53.3% to 100%, and population density was between 12.0 J2 to 85.0 J2 in 100ml soil. The result of different population density from different variety indicated that resistance to root-knot nematode was different among varieties.At greenhouse, results of assay on suitable condition for artifitional inoculatuin showed that inoculating amount of nemotode was 500 eggs per plant or 1 000 J2 per plant, pH of substrate was between pH5 to pH8, relative humidity of substrate was between 50% to 70%, temperature of the greenhouse were 20°C to 30°C, growth stage of banana seedling was at 6 leaves, investigation duration was 40 days to 60 days after inoculation, inoculating depth was 0 cm to 15 cm below surface of soil, texture of soil was sandy soil or mixture of clay soil and sandy soil(1:1).2 slightly susceptible varieties, 3 middle susceptible varieties, and one highly susceptible variety were found from field investigation. 48 banana germplasm were collected. Resistance of 42 germplasm was identificated with artificial inoculation method at greenhouse. Result indicated that one germplasm was middle resistant, 6 slightly susceptible, 16 middle susceptible , and 19 highly susceptible.
