The Trial studied the culture conditions of teliospore , and defined the optimum conditions for the teliospore germination. A antagonistic strain which has good inhibition against pathogens activity was screened, identified as Streptomyces, and systemic studied morphological characteristics, fermentation culture and culture conditions.Through the study of biological characteristics of Sphacelotheca reiliana teliospore, results shows teliospore grows best when the pH is 7 and optimal temperature is 28℃,the mixture of formaldehyde and Potassium can promote germination of teliospore, the germination rate of teliospore is add up to 84.49% on maltose-WA culture. Initially colony of Pathogen transforms from pre-yellow-white small point to Leather-like ,finally,the color became dark brown,dry surface, not smooth. Small teliopore is colorless, single-cell, elliptic, liquid of fermentation is reddish-brown..47 species of actinomyces and 41 species of bacteria were screened from soil against Sphace-lotheca reiliana, and screened strain SF-1 which has good inhibition against pathogens activity. The inhibition rate of SF-1 against germination of teliospore was add to76.56%. According to watching morphology, physiological and biochemical characteristic of antagonistic strain SF-1 in a set of cultures that is from ISP, the result showed that antagonistic strain SF-1 is Streptomyces.Through screening of cultures of antagonistic strains SF-1, the results showed peptone is necessary nutrient synthesized antibiotics, which culture have not peptone could not synthesized antibiotics, mycelium dry weight is 10.82 mg/mL, Inhibition rate of germination of teliospore is 61.15%. Sucrose is the best carbon source, Inhibition rate of germination of teliospore is 78.23%, mycelium dry weight is 21.26 mg/mL. Adding different concentrations of CaCO3, results showed 1% CaCO3 in culture can inhibit germination of teliospore, inhibition rate is77.44%, mycelium dry weight is 18.77mg/mL. Adding different concentrations of NaCl, results showed 0.4% NaCl in culture can inhibit germination of teliospore ,inhibition rate is 77.49%, mycelium dry weight is 18.32mg/mL. Through screening of culture conditions of antagonistic strains SF-1, the results showed that when the inoculation is2%, the inhibition rate of germination of teliopore is highest, mycelium dry weight is lightest. When temperature is 32℃, metabolites of SF-1 and mycelium dry weight both largest. When speed to 180 rpm, inhibition of metabolites of SF-1 against teliopore germination, germination inhibition rate is80.24%, mycelium dry weight is19.84 mg/mL. When pH is 7, activity of metabolites of SF-1 to teliopore is highest.When the volume ferment media is 80mL, antagonism activity and mycelium dry weight both largest.And the best time for metabolites production is 96h. Adopt the method of soaking seeds of maize determined the effect of SF-1 on the seeds germination and seedlings growth, found SF-1 has no risk to maize, could improve germination of the maize seeds, and promote growth of radicle and germ, Increase R/T.
